Son serina\/treonina kinasas citos\u00f3licas que se expresan en numerosos tipos de c\u00e9lulas. La mayor\u00eda de las c\u00e9lulas expresan al menos dos isoformas de pkds y las diferentes isoformas de las pkds poseen funciones espec\u00edficas en el tr\u00e1fico vesicular. Las pkds como efectoras activadas por la v\u00eda diacilglicerol (dag) no solo regulan la fisi\u00f3n y la formaci\u00f3n de ves\u00edculas en el tgn, sino que tambi\u00e9n podr\u00edan estar regulando otros puntos diferentes de control del tr\u00e1fico vesicular posteriores a la fisi\u00f3n de ves\u00edculas del tgn, como la generaci\u00f3n de cuerpos multivesiculares maduros (mvbs), as\u00ed como su transporte, anclaje y fusi\u00f3n a la membrana plasm\u00e1tica. Hemos empleado construcciones de pkd1 y modelos celulares carentes de uno o dos miembros de la familia de pkd para estudiar la implicaci\u00f3n de las pkds en diferentes puntos de control del tr\u00e1fico de mvbs  sobre los cuales la estimulaci\u00f3n celular y dgk alpha operan, focalizando el estudio en la maduraci\u00f3n de mvbs y el tr\u00e1fico de los mismos, que est\u00e1n implicados en la secreci\u00f3n de exosomas. La modulaci\u00f3n de la diacilglicerol kinasa alpha (dgk alpha) regula la activaci\u00f3n de pkd1 y su localizaci\u00f3n subcelular y estudios recientes se\u00f1alan a dgk alpha como un importante regulador de la generaci\u00f3n de cuerpos multivesiculares (mvbs) y secreci\u00f3n de exosomas en linfocitos t. La secreci\u00f3n de exosomas est\u00e1 alterada en c\u00e9lulas que expresan el dominante negativo de pkd1 (pkd1 kd) y aumenta cuando se expresa la pkd1 constitutivamente activa (pkd1 ca). En blastos t procedentes de ratones carentes de pkd2 la secreci\u00f3n de exosomas est\u00e1 disminuida y esto est\u00e1 asociado con la formaci\u00f3n de mvbs inmaduros. Se observ\u00f3 un comportamiento parecido en linfocitos b dt40 carentes de pkd1\/3.
