As\u00ed, se puede destacar el \u00e9xito de indicadores para calcio como el fura-2. Las gfps, dentro de su amplio uso en biolog\u00eda celular, han revolucionado el campo de los indicadores fluorescentes, permitiendo construir sondas recombinantes que se introducen en las c\u00e9lulas mediante transfecci\u00f3n y que permiten generar organismos transg\u00e9nicos. De esta forma se pueden construir sensores para el estudio de procesos celulares basados en diferentes t\u00e9cnicas de detecci\u00f3n, entre las que destaca fret y las gfps permutadas circularmente. en esta tesis abordamos el dise\u00f1o y caracterizaci\u00f3n de indicadores fluorescentes como herramienta de estudio de distintos procesos que ocurren en el interior de las c\u00e9lulas vivas. Con ayuda de las bien conocidas prote\u00ednas fluorescentes verdes (gfps, green fluorescent proteins) y el fen\u00f3meno de transferencia de energ\u00eda por resonancia de fluorescencia (fret, fluorescence resonance energy transfer) hemos construido sensores para el estudio de receptores nucleares: receptor de estr\u00f3geno (er, estrogen receptor) y receptor de progesterona (pr, progesterone receptor). Adem\u00e1s, como alternativa a estos indicadores basados en fret, hemos desarrollado sensores para la detecci\u00f3n de ligandos de er basados en prote\u00ednas fluorescentes permutadas circularmente, en las cuales cambian las caracter\u00edsticas espectrales dependiendo de su estado de protonaci\u00f3n. Tambi\u00e9n hemos construido sensores para la detecci\u00f3n de actividad de la proteasa ns3\/4a del virus de la hepatitis c (hcv, hepatitis c virus). este tipo de sensores podr\u00edan ser utilizados para identificar sustancias con capacidad de uni\u00f3n a er y pr en librer\u00edas de compuestos en la industria farmac\u00e9utica, y permitir\u00eda caracterizar la afinidad ligando-receptor. Los indicadores de actividad protesasa de hcv servir\u00e1n tanto para la b\u00fasqueda de inhibidores  para terapia antiviral, como para la caracterizaci\u00f3n de variantes de los distintos
