En modelos experimentales de diabetes la expresi\u00f3n renal del glut-1 est\u00e1 aumentada. Por otro lado, el sistema-renina-angiotensina-aldosterona (sraa), est\u00e1 estrechamente relacionado con la nefropat\u00eda diab\u00e9tica (nd). La enzima convertidora de angiotensina-2 (eca-2), es la principal enzima degradadora de angiotensina ii (ang-ii). En animales diab\u00e9tico y en biopsias renales de humanos diab\u00e9ticos, la expresi\u00f3n glomerular de la eca2 esta disminuida. Nosotros postulamos que la sobrexpresi\u00f3n del glut-1 podr\u00eda contribuir a una hiperactivaci\u00f3n del sraa, mediante la alteraci\u00f3n de la eca-2.  objetivo: valorar los cambios histol\u00f3gicos presentes en un modelo murino transg\u00e9nico que sobrexpresa el transportador glut-1. Estudiar la expresi\u00f3n de las enzimas eca y eca2 tanto en el modelo animal como in vitro en c\u00e9lulas mesangiales que sobrexpresan el transportador glut-1. M\u00e9todos: para realizar los estudios en vivo e in vitro utilizamos modelos transg\u00e9nicos animales y de c\u00e9lulas mesangiales que sobrexpresan el transportador de glucosa glut-1. La expresi\u00f3n proteica de las enzimas eca y eca2 se valorar\u00f3 con inmunohistoqu\u00edmica, inmunofluerescencia y t\u00e9cnicas de western-blot. La expresi\u00f3n de arnm se estudiar\u00f3 mediante t\u00e9cnicas de reacci\u00f3n en cadena de la polimerasa en tiempo real (pcr real time). La actividad de los enzimas fue medida mediante ensayo de apagado de sustrato fluorescente. Resultados: los animales transg\u00e9nicos mostraron  signos de glomerulopat\u00eda diab\u00e9tica acompa\u00f1ado de un aumento en la expresi\u00f3n glomerular de la eca y disminuci\u00f3n de la eca2. Los estudios in vitro revelaron que la sobrexpresi\u00f3n del glut-1 aumentaba la expresi\u00f3n transcripcional de la eca y disminu\u00eda la expresi\u00f3n tanto transcripcional como post-transcripcional de la eca2. Nuestros resultados ponen de manifiesto el importante papel que juega el glut-1 en el desarrollo de la nd y en la activaci\u00f3n del sraa.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
