En este sistema la importina alfa reconoce una se\u00f1al de localizaci\u00f3n nuclear (nls) en la prote\u00edna cargo y la importina beta interacciona con componentes del complejo de poro nuclear para permitir la translocaci\u00f3n del complejo a trav\u00e9s de la envoltura nuclear. La nucleoplasmina (np), un chaper\u00f3n de histonas nuclear homopentam\u00e9rico, presenta el prototipo de nls cl\u00e1sica bipartita en cada uno de los dominios c-terminales (cola de sus mon\u00f3meros). En esta tesis hemos caracterizado la formaci\u00f3n de un complejo de np saturado con 5 mol\u00e9culas de importina alfa, as\u00ed como los par\u00e1metros termodin\u00e1micos que regulan dicho reconocimiento.   adem\u00e1s, presentamos un modelo 3d a baja resoluci\u00f3n, obtenido por dispersi\u00f3n de rayos x a bajo \u00e1ngulo, en el que se describe por primera vez, la forma y orientaci\u00f3n relativa de las prote\u00ednas en un complejo de importaci\u00f3n. la nucleoplasmina, que interviene en procesos de remodelaci\u00f3n de la cromatina, interacciona con histonas: act\u00faa como reservorio de histonas nucleos\u00f3micas h2a\/h2b, y tambi\u00e9n se une a histonas de tipo conector. Los dominios cola de la nucleoplasmina est\u00e1n involucrados en la interacci\u00f3n con histonas. Dado que las secuencias nls tambi\u00e9n se localizan en los dominios cola, hemos estudiado como la presencia de histonas (nucleos\u00f3micas y de tipo conector) afecta al reconocimiento de np por importina alfa. Analizamos la formaci\u00f3n de complejos ternarios np\/histonas\/importina alfa, cuya existencia sugiere un posible mecanismo de cotransporte mediado por la importina
