La prote\u00edna de fusi\u00f3n ews\/fli1 es un factor de transcripci\u00f3n aberrante con propiedades oncog\u00e9nicas y esencial para el desarrollo de los tumores de ewing. Nuestro grupo ha demostrado que ews\/fli1 induce la expresi\u00f3n de los genes cck y nr0b1, y que reprime la expresi\u00f3n del gen lox (lisil oxidasa). Profundizamos en el mecanismo de regulaci\u00f3n de lox por parte de ews\/fli1 y estudiamos su papel en los tumores de ewing. Para ello desarrollamos sistemas inducibles de expresi\u00f3n de lox, as\u00ed como de su regi\u00f3n prop\u00e9ptido (pp-lox) y de la enzima madura (lox enz). De esta manera demostramos que la inducci\u00f3n de la regi\u00f3n prop\u00e9ptido de lox en l\u00edneas celulares de ewing bloquea el ciclo celular, disminuye la proliferaci\u00f3n celular y la capacidad para formar tumores en ratones inmunodeprimidos, mientras que observamos los efectos contrarios al sobreexpresar lox enzima. Nuestros resultados indican que en los tumores de ewing, la regi\u00f3n prop\u00e9ptido de lox act\u00faa como supresor tumoral y que por tanto su expresi\u00f3n o administraci\u00f3n podr\u00eda ser una herramienta para el tratamiento de este tumor pedi\u00e1trico tan agresivo. summary: ewing family of tumors are characterized by the presence of traslocations between the ews gen and some members of the ets family of transcription factors, being fli1 the most frequently implicated in the traslocation. The fusion protein ews\/fli1 is an aberrant transcription factor with oncogenic properties and essential for ewing tumor development. Our group has demonstrated that ews\/fli1 induces cck and nr0b1 expression and reprimes lox expression. We then focus in the regulation mechanism of lox by ews\/fli1, and in its role in ewing tumors. We use inducible models to express lox, the propeptide region and the mature enzyme expression. In this way we demonstrate that the induction of the propeptide region of lox in ewing cell lines causes growth arrest, reduces cellular proliferation and the transforming capability to growth in immunodeficient mice. The expression of lox enzyme has the contrary effect. Our data indicates that the propeptide region of lox acts as a tumor suppressor in ewing tumors, so its expression or administration could be a new approach in the treatment of this aggressive
