Consiste en la digestion del dna a analizar con la restrictasa adecuada, de forma que las moleculas silvestres se cortaran y las mutantes, que quedan intactas seran amplificadas por pcr.  experimentos de reconstruccion con plasmidos mostraron que la sensibilidad del ensayo era de una molecula mutante entre 10.000.000 de copias silvestres. No todas las restrictasas analizadas mostraron la eficiencia necesaria para su uso en rsm. Los problemas de contaminacion cruzada se resolvieron aislando las zonas pre y postpcr. Se realizaron ensayos in vivo con bacterias silvestres o deficientes en alquil transferasas. Se comprobo una sensibilidad de una secuencia mutante entre 100.000 genomas, pudiendose observar la progresion de los procesos de reparacion del da\u00f1o en el dna. Experimentos con celulas humanas mostraron una sensibilidad similar, asi como la reparacion del da\u00f1o.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
