La sangre de cord\u00f3n umbilical (scu) representa una fuente adecuada para la obtenci\u00f3n de hscs dado que proporciona diversas ventajas en comparaci\u00f3n con la m\u00e9dula \u00f3sea o la sangre perif\u00e9rica movilizada. Sin embargo, posee una serie de limitaciones como el bajo n\u00famero total de hscs o el retraso en el tiempo de recuperaci\u00f3n de diversos linajes hematopoy\u00e9ticos tras el transplante. Por esos motivos, resulta necesario profundizar en el conocimiento de los mecanismos celulares y moleculares que regulan la autorrenovaci\u00f3n y diferenciaci\u00f3n de las hscs, adem\u00e1s de ahondar en la b\u00fasqueda y desarrollo de nuevos sistemas in vitro que permitan mantener y expandir dicha poblaci\u00f3n.  \tpor ello, en el presente trabajo se trat\u00f3 de aislar y caracterizar las hscs en base al marcador cd34, para posteriormente analizar las poblaciones hematopoy\u00e9ticas diferenciadas generadas durante la expansi\u00f3n in vitro de dichas c\u00e9lulas cd34+. Una vez caracterizadas todas las poblaciones en estudio en base a marcadores de membrana, procedimos a estudiar el efecto de diferentes factores de crecimiento, inhibidores de la maquinaria epigen\u00e9tica y c\u00e9lulas de soporte sobre el mantenimiento, expansi\u00f3n y diferenciaci\u00f3n de las hscs. Adem\u00e1s, se realiz\u00f3 un an\u00e1lisis de expresi\u00f3n g\u00e9nica bajo condiciones de cultivo controladas, as\u00ed como una b\u00fasqueda bibliogr\u00e1fica de otros an\u00e1lisis de expresi\u00f3n g\u00e9nica en poblaciones de c\u00e9lulas madre con el fin de encontrar genes candidatos a participar en el proceso de autorrenovaci\u00f3n de las hscs. Finalmente, se analiz\u00f3 el efecto de algunos de estos genes candidatos sobre la autorrenovaci\u00f3n y diferenciaci\u00f3n de hscs mediante el uso de vectores lentivirales para la inhibici\u00f3n
