Est\u00e1 implicada en numerosos procesos clave, tales como la proliferaci\u00f3n, la diferenciaci\u00f3n, la apoptosis, la migraci\u00f3n, y la tumorog\u00e9nesis. La prote\u00edna kinase suppressor of ras 1 (ksr1) es esencial en el control de esta v\u00eda de se\u00f1alizaci\u00f3n, al funcionar como un scaffold molecular sobre el que se ensamblan los distintos componentes individuales del signalosoma raf\/mek\/erk. la funci\u00f3n scaffold de ksr1 est\u00e1 regulada principalmente por las prote\u00ednas 14-3-3. \u00e9stas constituyen una gran familia de prote\u00ednas altamente conservadas que se expresan ubicuamente en las c\u00e9lulas eucariotas. En mam\u00edferos se han descrito siete isoformas, codificadas por genes distintos y denominadas p y, e, tj, o, t y estas prote\u00ednas forman d\u00edmeros capaces de unir motivos consenso que contienen residuos serina y\/o treonina fosforilados. En concreto, ksr1 interacciona con 14-3-3 a trav\u00e9s de los residuos s297 y s392, mediante un mecanismo regulado por la gtpasa ras. Existen pocos trabajos que hayan investigado las funciones espec\u00edficas de las isoformas 14-3-3 y, en particular, se desconoc\u00eda si ksr1 interacciona de manera promiscua con todas las isoformas 14-3-3 o, por el contrario, la asociaci\u00f3n ocurre de manera selectiva, con determinadas isoformas. utilizando diferentes aproximaciones experimentales, hemos demostrado en esta memoria que, in vivo, ksr1 interacciona espec\u00edficamente con las isoformas 14-3-3y y 14-3-3t). Esta interacci\u00f3n es funcionalmente relevante ya que regula la localizaci\u00f3n subcelular de ksr1 y su funci\u00f3n scaffold para erk. Adem\u00e1s, hemos demostrado que el segmento variable carboxi-terminal de 14-3-3 confiere especificidad a la interacci\u00f3n con ksr1 y que la fracci\u00f3n de 14-3-3y unida a ksr1 forma homod\u00edmeros y heterod\u00edmeros \u00fanicamente con las isoformas 14-3-3t y 14-3-3^. En resumen, nuestros datos indican que, a pesar de compartir los mismos motivos consenso de uni\u00f3n, existen determinantes moleculares, tanto en las isoformas 14-3-3 como en los ligandos, que
