En este trabajo se ha estudiado el papel del proteasoma en la proteostasis de a-sinucle\u00edna. La sobreexpresi\u00f3n de a-sinucle\u00edna no tiene un efecto significativo sobre la expresi\u00f3n, ensamblaje y maduraci\u00f3n de las subunidades del proteasoma para la formaci\u00f3n del complejo activo,  y tampoco afecta las actividades peptidasa del proteasoma. A-sinucle\u00edna interacciona in vitro con la subunidad c8 (a7) del proteasoma 20s a trav\u00e9s de su regi\u00f3n amino-terminal, y el an\u00e1lisis de los p\u00e9ptidos resultantes de su degradaci\u00f3n indica que la digesti\u00f3n inicial tiene lugar en el extremo amino-terminal. La oxidaci\u00f3n de las metioninas  1 y 5 del n-terminal de a-sinucle\u00edna inhibe su degradaci\u00f3n por el proteasoma, y esta inhibici\u00f3n es reversible mediante su reparaci\u00f3n por la metionina sulforeductasa a. Sinfilina-1 inhibe la degradaci\u00f3n de a-sinucle\u00edna por el proteasoma, y esta inhibici\u00f3n se revierte por coexpresi\u00f3n de siah-1 que promueve la ubicuitilaci\u00f3n y degradaci\u00f3n de sinfilina-1. La fosforilaci\u00f3n de a-sinucle\u00edna en ambas regiones amino y carboxilo terminales de la prote\u00edna inhibe su degradaci\u00f3n por el proteasoma, independientemente de las quinasas participantes en dicha fosforilaci\u00f3n.
