Para evitar encapsidaci\u00f3n del virus helper, su se\u00f1al de empaquetamiento (&#936;) est\u00e1 flanqueada por secuencias de reconocimiento de recombinasas expresadas por las c\u00e9lulas empaquetadoras. Sin embargo, frecuentemente se observa una acumulaci\u00f3n de virus helper y un bajo rendimiento en la producci\u00f3n de hc-ad. Esto es debido, en parte, a la expresi\u00f3n insuficiente de la recombinasa por parte de las c\u00e9lulas empaquetadoras. En el presente trabajo se describe un nuevo virus helper (adtetcre) donde su secuencia &#936; est\u00e1 flanqueada por sitios loxp que pueden ser escindidos por una recombinasa quim\u00e9rica llamada mercremer codificada por el genoma del propio virus. La modulaci\u00f3n eficiente de la escisi\u00f3n se logra con un control simult\u00e1neo de la expresi\u00f3n mercremer utilizando un sistema inducible tet-on y su translocaci\u00f3n al n\u00facleo por 4-hidroxitamoxifeno. La encapsidaci\u00f3n del virus adtetcre se inhibe mediante doxiciclina y 4-hidroxitamoxifeno. Para la producci\u00f3n del hc-ad se utilizan c\u00e9lulas 293cre4 y el virus adtetcre donde se observa que existe una cooperaci\u00f3n entre la recombinasa celular y la codificada por el virus minimizando la contaminaci\u00f3n con virus helper. Este nuevo m\u00e9todo es altamente reproducible y permite la producci\u00f3n rutinaria en el laboratorio de diferentes hc-ad a mediana escala con c\u00e9lulas adherentes, obteniendo t\u00edtulos mayores de 1010 unidades infecciosas y menos del 0,1% de contaminaci\u00f3n de virus helper. La contaminaci\u00f3n residual de virus helper adtetcre carece de secuencia &#936; y est\u00e1n altamente atenuados. Como conclusi\u00f3n, el virus helper auto-inactivable adtetcre es una herramienta prometedora para la producci\u00f3n del hc-ad.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
