Carmen <\/strong><\/h2>\n<p>El trabajo presentado describe el desarrollo de un sistema de transformacion estable para t. Cruzi, que permite la integracion dirigida en el genoma del parasito de genes foraneos (hph, neo, luc), utilizando la diana genica 1f8 como lugar de insercion.  la integracion se produce por doble recombinacion homologa entre las secuencias del gen 1f8 portadas por los diversos vectores y sus homologas presentes en el genoma de t. Cruzi. La secuenciacion del cdna correspondiente al mrna de los transformantes yhph1f8 evidencia que la recombinacion se produce a nivel de nucleotido.  las se\u00f1ales de procesamiento que se ligaron a los genes foraneos, se\u00f1ales de trans-splicing y putativa de poliadenilacion de los genes 1f8 y kap, para la formacion de mrnas traducibles, fueron utilizadas por la maquinaria del splicing para la maduracion de los pre-mrnas de los genes hph y luc resultando, en los transformantes yhph1f8 y yhphluc, mensajeros poliadenilados de 1300 y 2200 nucleotidos respectivamente. Estos tama\u00f1os corresponden con lo esperado, tras la utilizacion de las se\u00f1ales de aceptor de splicing y poliadenilacion introducidas.  la introduccion de secuencias reguladoras de la expresion genica (aceptor de splicing y poliadenilacion) pertenecientes a genes estadio especifico, condiciona su expresion a dichos estadios. Asi los transformantes yhphluc, que contienen insertadas dichas secuencias, expresan altos niveles de luciferasa en las formas no infectivas pero no en las formas infectivas. Los transformantes yhphplgp72luc han expresado luciferasa en menor cantidad que los transformantes yhphluc.  la eliminacion del gen mip, mediante co-transformacion o transformaciones sucesivas con varios vectores, no ha sido posible probablemente por resultar su ausencia letal para t.
