As\u00ed, toda una serie de evidencias acumuladas durante los \u00faltimos a\u00f1os en diferen tes modelos experimentales indican que el ?9-tetrahydrocannabinol (thc), el principal constituyente de la marihuana y otros agonistas de los receptores de cannabinoides inhiben el crecimiento de diferentes tipos de tumores, incluyendo los gliomas. Un a de las caracter\u00edsticas de mayor relevancia cl\u00ednica del glioblastoma multiforme (gbm) es su extrema resistencia a los tratamientos convencionales. Esta caracter\u00edstica deriva entre otros factores de la alta malignidad y heterogeneidad de las c\u00e9lulas que consitiyen esste tipo de tumores. Por tanto, es crucial desarrollar nuevas estrategias terap\u00e9uticas dirigidas a mejorar el pron\u00f3stico de los pacientes que sufren esta patolog\u00eda.   con estos antecedentes, y dado que era esperable que muchos tumore s presentasen resistencia al tratamiento con cannabinoides, en la presente tesis doctoral quisimos responder a dos cuestiones cuya respuesta podr\u00eda facilitar la optimizaci\u00f3n de una terapia antitumoral basada en la utilizaci\u00f3n de cannabinoides: 1) \u00c2\u00bfes  posible predecir en base a sus caracter\u00edsticas moleculares si un determinado tumor es sensible al tratamiento con estos compuestos? Y 2) \u00c2\u00bfpuede potenciarse la acci\u00f3n de los cannabinoides por la administraci\u00f3n combinada de otros f\u00e1rmacos antitumorale s?.  as\u00ed por un lado, hemos identificado una serie de genes espec\u00edficamente asociados con la resistencia al thc mediante el an\u00e1lisis del perfil de la expresi\u00f3n g\u00e9nica de un conjunto de c\u00e9lulas de glioma humano con diferente sensibilidad a la muerte c elular inducida por cannabinoides. Adem\u00e1s, hemos encontrado que uno de estos genes, el factor de crecimiento midquina (mdk) esta directamente involucrado en la resistencia de las c\u00e9lulas de glioma al tratamiento con thc. En este estudio, hemos mostra do que la mdk media su efecto protector a trav\u00e9s del receptor tirosina quinasa alk (anaplasic lymphoma kinase receptor), y que la se\u00f1alizaci\u00f3n de alk inducida por mdk impide la activaci\u00f3n de la ruta de muerte autof\u00e1gica inducida por los cannabinoides .
