Las neurotrofinas intervienen en la mayor\u00eda de los procesos del neurodesarrollo (proliferaci\u00f3n, neurog\u00e9nesis, neuritog\u00e9nesis, plasticidad sin\u00e1ptica, supervivencia celular, migraci\u00f3n, etc.) Y en la funcionalidad del cerebro adulto y, por tanto ser\u00edan buenos candidatos a participar como factores predisponentes en diferentes enfermedades mentales. Estudios previos de nuestro grupo han sugerido la implicaci\u00f3n del receptor de la neurotrofina 3, trkc, en el trastorno de p\u00e1nico. En el contexto de esta tesis, proponemos que la sobrexpresi\u00f3n del gen ntrk3 (trkc) es un mediador com\u00fan de los desencadenantes gen\u00e9ticos y ambientales en este trastorno. Concretamente, creemos que su deregulaci\u00f3n podr\u00eda producir cambios estructurales y funcionales en la corteza cerebral de los pacientes debido a su papel en el establecimiento de la circuiter\u00eda cortical y la neuroplasticidad en el adulto. En concreto, estas alteraciones en el desarrollo y la funci\u00f3n corticales podr\u00edan ser elementos de predisposici\u00f3n a sufrir ataques de p\u00e1nico en los pacientes que padecen esta enfermedad.  nuestro trabajo se ha centrado en la estructura de la corteza cerebral y en el proceso de corticog\u00e9nesis y neuritog\u00e9nesis de un modelo experimental de trastorno de p\u00e1nico. As\u00ed pues, los objetivos principales de este trabajo han sido: 1\/ determinar la contribuci\u00f3n espec\u00edfica del gen ntrk3 en las alteraciones de la corteza cerebral observadas en los pacientes con el trastorno de p\u00e1nico, mediante el uso de un modelo murino modificado gen\u00e9ticamente (tgntrk3), y 2\/ analizar el impacto espec\u00edfico de la sobrexpresi\u00f3n de ntrk3 sobre la corticog\u00e9nesis durante estad\u00edos embrionarios o postnatales estudiando los procesos de generaci\u00f3n y neuritog\u00e9nesis de las neuronas corticales. los resultados obtenidos indican que la sobrexpresi\u00f3n de ntrk3 en ratones da lugar a una reducci\u00f3n del grosor del l\u00f3bulo frontal, reproduciendo en nuestro modelo la hipofrontalidad observada en los pacientes con trastorno de p\u00e1nico que conllevar\u00eda una menor inhibici\u00f3n de los n\u00facleos subcorticales del sistema l\u00edmbico, como la am\u00edgdala. Se observan adem\u00e1s una serie de alteraciones citoarquitect\u00f3nicas en la corteza prefrontal medial que apoyan la hip\u00f3tesis de la malfunci\u00f3n de esta regi\u00f3n. Adem\u00e1s, los ratones tgntrk3 presentan cambios estructurales en la corteza somatosensorial primaria, sugiriendo que el procesamiento de la informaci\u00f3n sensorial podr\u00eda ser en cierta medida aberrante disfuncionalidad cortical hasta ahora no explorada en pacientes. Estos cambios observados en el adulto podr\u00edan depender de cambios en la corticog\u00e9nesis en estad\u00edos postnatales debidos a la sobrexpresi\u00f3n de ntrk3, que afectan tambi\u00e9n a la neuritog\u00e9nesis en cultivos primarios corticales, adem\u00e1s de modificar la respuesta de las neuronas a la estimulaci\u00f3n con neurotrofinas.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de
