Esther <\/strong><\/h2>\n<p>Se han purificado dos formas isoenzimaticas de la isocitrato deshidrogenasa dependiente de nad (idh i e idh ii) de micelio de phycomyces blakesleeanus nrrl 1555(-), con un rendimiento proximo al 10%. De las diferencias obtenidas entre las dos isoenzimas en el estudio de parametros fisico-quimicos, se ha estimado que la idh ii posee una carga neta negativa superior a la que posee la idh i.  la forma activa del isocitrato es la tribasica para las dos isoenzimas. A ph 7,6, el mecanismo cinetico permite la formacion del complejo activo enzima-magnesio-isocitrato mediante dos vias alternativas, siendo la concentracion de mg2+ la que determina el proceso predominante. Se obtuvo cinetica sigmoide para la forma activa del isocitrato como sustrato, con diferentes valores de nh para las dos isoenzimas. Para el nad se obtuvo cinetica de michaelis-menten. El citrato es un activador alosterico para las dos isoenzimas, y el amp lo es para la idh i.
