En este contexto, las prote\u00ednas long coiled-coil participan en varias etapas de estos procesos, actuando bien como factores de tethering en el tr\u00e1fico intracelular de membranas o como factores de scaffolding de redes moleculares complejas. Estudios previos en recientes de nuestro laboratorio han permitido la identificaci\u00f3n de un gen espec\u00edfico de vertebrados denominado necc1 (neuroendocrine long coiled-coil protein 1), sobre la base de sus caracter\u00edsticas estructurales y su expresi\u00f3n preferencial en tejidos nerviosos y \u00f3rganos endocrinos. Necc1 pertenece a una familia de genes conformada por dos miembros adicionales denominados jakmip1\/marlin1 y necc2 (neuroendocrine long coiled-coil protein 2), presentando \u00e9ste \u00faltimo homolog\u00eda estructural y expresi\u00f3n preferencial en los mismos tejidos que necc1, aun cuando su funci\u00f3n espec\u00edfica est\u00e1 por descubrir. En este sentido, el objetivo general de la tesis consiste en la caracterizaci\u00f3n bioqu\u00edmica, celular y funcional de la prote\u00edna necc2 mediante el an\u00e1lisis de su distribuci\u00f3n y de los mecanismos moleculares a trav\u00e9s de los cuales esta prote\u00edna puede estar implicada en el funcionamiento de procesos relacionados con el tr\u00e1fico intracelular de membranas y la se\u00f1alizaci\u00f3n en c\u00e9lulas neuroendocrinas.  \tlos resultados principales obtenidos en el presente estudio son los siguientes: necc2 constituye una prote\u00edna transmembrana cuyo anclaje en membranas celulares se produce a trav\u00e9s de su regi\u00f3n hidrof\u00f3bica carboxilo-terminal. Adicionalmente, las regiones coiled-coil de la prote\u00edna necc2 son responsables de su asociaci\u00f3n en olig\u00f3meros de alto peso molecular, caracter\u00edstica particular en este tipo de prote\u00ednas. en c\u00e9lulas pc12, necc2 se localiza en caveolas, donde colocaliza con caveolina-1 y con el receptor de neurotrofinas trka. Adicionalmente, la expresi\u00f3n y distribuci\u00f3n de necc2 en estas c\u00e9lulas est\u00e1 regulada por el ligando de dicho receptor, ngf. M\u00e1s a\u00fan, necc2 sufre splicing alternativo en este modelo celular, lo cual regula su localizaci\u00f3n y el tipo de asociaci\u00f3n que mantiene con las membranas celulares. la sobreexpresi\u00f3n y\/o el silenciamiento de necc2 modifican la fosforilaci\u00f3n de un importante mediador  intracelular asociado a la activaci\u00f3n del receptor trka, erk1\/2. en conjunto, los resultados anteriores sugieren que necc2 podr\u00eda participar como prote\u00edna de scaffolding o tethering vesicular, modulando la funcionalidad de caveolas y del receptor trka en c\u00e9lulas
