Los ratones knock out de p53 son propensos a desarrollar distintos tipos tumorales a edades tempranas; sin embargo, no se sabe cu\u00e1l es el per\u00edodo en el que la falta de p53 es necesaria para el desarrollo de un tumor o si \u00e9ste, una vez establecido, podr\u00eda eliminarse reactivando a p53.   el rat\u00f3n knock-in p53ertamki (christophorou et al, 2005) permite un control temporal y reversible de la funci\u00f3n de p53 mediante la inyecci\u00f3n de tamoxif\u00e9n. Se ha utilizado este modelo para estudiar la posible cooperaci\u00f3n entre la falta de funci\u00f3n de p53 y la prote\u00edna oncog\u00e9nica p210bcr-abl, responsable de la leucemia mieloide cr\u00f3nica en humanos. Para ello hemos cruzado ratones p53ertamki con transg\u00e9nicos que expresan el oncog\u00e9n p210bcr-abl en el compartimento stem (perez-caro et al, 2009) obteniendo ratones de diversos genotipos, en los cuales hemos estudiado la supervivencia y la progresi\u00f3n de la enfermedad. Nuestros datos muestran que la falta de funci\u00f3n de p53 acelera el proceso tumoral causado por el oncog\u00e9n. Tambi\u00e9n hemos observado que la restauraci\u00f3n de la funci\u00f3n de p53 durante tan solo una semana es suficiente para mejorar la supervivencia de estos ratones. Nuestros resultados sugieren que el proceso desencadenado por p53 para frenar el progreso de la enfermedad es el de muerte celular o apoptosis de los progenitores hematopoy\u00e9ticos o c\u00e9lulas sca1+.
