El primero, incluye trabajo de investigaci\u00f3n dedicado a la identificaci\u00f3n de nuevos inhibidores selectivos de la pop en extractos de plantas y el segundo en experimentos basados en la resonancia magn\u00e9tica nuclear para realizar estudios estructurales de pop que incluyen su din\u00e1mica conformacional en soluci\u00f3n e interacci\u00f3nes prote\u00edna ligando.  \ten una primera parte y sabiendo que se han identificado pocos inhibidores de la pop con esqueletos diferentes a los pept\u00eddicos se estableci\u00f3 como objetivo la identificaci\u00f3n de mol\u00e9culas activas en extractos acuosos de plantas medicinales usadas en la herbolaria en china y, en un trabajo en colaboraci\u00f3n con la dra. Ionara dalcol, en planas brasile\u00f1as. Este trabajo de investigaci\u00f3n resulto en la identificaci\u00f3n de seis nuevos productos naturales con actividad inhibidora de pop: berberina, bacalina, hispidulina, aroxylosida y lupeol. \t \ten la segunda parte de esta tesis se  han realizado estudios de la pop basados en la resonancia magn\u00e9tica nuclear como t\u00e9cnica principal de observaci\u00f3n y an\u00e1lisis. Primero, se ha desarrollado un m\u00e9todo de cribado de inhibidores selectivos de pop y dpp iv basado en la resonancia de fl\u00faor y se aplico para la identificaci\u00f3n de actividad inhibidora en extractos acuosos de plantas.   adem\u00e1s se ha desarrollado una metodolog\u00eda para el estudio de biomol\u00e9culas de alto peso molecular por resonancia magn\u00e9tica nuclear. Este metodo consiste en el marcaje isotopico selectivo con 15n de las cadenas laterales de los tript\u00f1ofanos, por medio de la adici\u00f3n de indol enriquecido con 15n al sistema de expresi\u00f3n de la prote\u00edna en medio m\u00ednimo. A continuaci\u00f3n se demostr\u00f3 la fiabilidad de este metodo con la prolil oligopeptidase y se estudio la interaccion con un inhibidor, zpp. En otro trabajo de investigaci\u00f3n se utiliz\u00f3 este m\u00e9todo en combinaci\u00f3n con m\u00e9todos modernos de resonancia magn\u00e9tica nuclear y de expresi\u00f3n de prote\u00ednas en e. Coli para estudiar la din\u00e1mica conformacional de pop en soluci\u00f3n. En concreto, se produjo una muestra altamente deuterada y uniformemente marcada con 15n de pop y se aplico un experimento basado en el an\u00e1lisis del tiempo de relajaci\u00f3n transversal de la se\u00f1al de 15n por resonancia magn\u00e9tica nuclear y se correlacionaron los resultados con la estructura de pop utilizando el marcaje selectivo de tript\u00f3fano.
