La sobreexpresi\u00f3n de e2f1 induce apoptosis en varias poblaciones neuronales; y niveles end\u00f3genos de e2f1 se encuentran elevados en enfermedades neurodegenerativas como el alzheimer y la enfermedad de parkinson, que se caracterizan por un elevado \u00edndice en la muerte de c\u00e9lulas neuronales. En este trabajo se propuso estudiar los efectos apopt\u00f3ticos de e2f1 y divulgar los mecanismos de muerte celular promovidos por la sobreexpresi\u00f3n de e2f1 en un modelo de c\u00e9lulas neuronales. Este estudio demuestra que la sobreexpresi\u00f3n de e2f1 induce la apoptosis en c\u00e9lulas pc12 privadas de suero, en el estado nativo y diferenciado, y que la acci\u00f3n apopt\u00f3tica de e2f1 en ambos estadios requiere de la actividad de gsk3-beta. Nuestros resultados sugieren que la actividad de gsk3-beta interfiere con la actividad transcripcional de e2f1 y por este mecanismo gsk3-beta podr\u00eda estar modulando la expresi\u00f3n de genes diana de e2f1. La funci\u00f3n apopt\u00f3tica de e2f1 se produce por la v\u00eda de se\u00f1alizaci\u00f3n intrinsica, y implica la regulaci\u00f3n positiva de biml y la activaci\u00f3n y translocaci\u00f3n de bax a la mitocondria, junto con una disminuci\u00f3n del bcl-xl mitocondrial. E2f1 es tambi\u00e9n capaz de inducir la acumulaci\u00f3n de ros en nuestro modelo celular. El antioxidante n-acetilciste\u00edna inhibi\u00f3 la translocaci\u00f3n de bax a la mitocondria y, en consecuencia, la apoptosis inducida por e2f1. El tratamiento de las c\u00e9lulas con diphenyleneiodonium, un inhibidor de la nadph oxidasas, no fue capaz de reducir totalmente la acumulaci\u00f3n de ros inducida por e2f1 y el efecto apoptotico de e2f1 se sigui\u00f3 observando en este caso. Por otra parte, e2f1 ha sido capaz de inhibir la actividad de nf-kb, lo que sugiere que las enzimas diana de nf-kb con funciones en la regulaci\u00f3n de ros podr\u00edan ser las responsables de los efectos de e2f1 en la producci\u00f3n de ros. La correlaci\u00f3n directa entre la funci\u00f3n apopt\u00f3tica de e2f1 y la producci\u00f3n de ros tambi\u00e9n se confirm\u00f3 en dos l\u00edneas celulares de neuroblastoma, sh-sy5y y sk-n-jd. La sobreexpresi\u00f3n de e2f1 en sh-sy5y indujo apoptosis, as\u00ed como la producci\u00f3n de ros; en cambio, la sobre-expresi\u00f3n de e2f1 en sk-n-jd no produjo ninguna alteraci\u00f3n en los niveles de ros y no indujo la apoptosis.
