Fern\u00e1ndez <\/strong><\/h2>\n<p>Gracias a las peculiaridades que presenta el adn mitocondrial (m\u00faltiples copias por c\u00e9lula, herencia materna, ausencia de recombinaci\u00f3n, alta tasa de mutaci\u00f3n), el an\u00e1lisis de su variabilidad pudo ser aplicado, en la presente tesis, a diferentes campos de estudio: en el campo de la gen\u00e9tica de poblaciones: permiti\u00f3 analizar el origen y la dispersi\u00f3n de las poblaciones humanas modernas en el campo de la gen\u00e9tica forense: permiti\u00f3 investigar muestras forenses degradadas o sin adn nuclear (como huesos y cabellos sin bulbo). en el campo de la gen\u00e9tica cl\u00ednica: permite la identificaci\u00f3n de posibles mutaciones que pueden causar una enfermedad o participar de ella; adem\u00e1s de detectar una posible estratificaci\u00f3n en los estudios caso-control que podr\u00eda suponer el establecimiento de una relaci\u00f3n err\u00f3neas entre una variante gen\u00e9tica y una enfermedad. en el presente proyecto de tesis, adem\u00e1s del desarrollo de un protocolo para la obtenci\u00f3n del genoma mitocondrial completo, se han desarrollado diferentes m\u00e9todos de minisecuenciaci\u00f3n, que consisten en la incorporaci\u00f3n de una sola base en una posici\u00f3n conocida que presenta variabilidad entre individuos, como son snapshot (applied biosystems, foster city, ca, usa) que detecta la variante por fluorescencia o por primera vez por medio de maldi-tof ms (sequenom, san diego, ca, usa) que detecta la variante por espectrometr\u00eda de
