Estudios metab\u00f3licos in vivo realizados previamente en portadores heterocigotos de esta variante de apo a-i revelaron que tiene una tasa de catabolismo que duplica la de la prote\u00edna nativa sugiriendo la posibilidad de que tuviera un efecto favorable sobre el proceso global de transporte reverso de colesterol. el objetivo de este trabajo fue establecer un sistema de expresi\u00f3n y purificaci\u00f3n de apo a-i nativa y apo a-i z recombinantes de alto rendimiento y elevada pureza con el fin de comparar in vitro sus propiedades en distintos aspectos relativos al transporte reverso de colesterol, as\u00ed como otras caracter\u00edsticas antiaterog\u00e9nicas de la apo a-i. se han optimizado la expresi\u00f3n y el m\u00e9todo de purificaci\u00f3n de la apo a-i nativa y apo a-i z adem\u00e1s de producir part\u00edculas reconstituidas similares a la hdl (rhdl) para poder realizar una caracterizaci\u00f3n estructural y funcional. la caracterizaci\u00f3n estructural incluy\u00f3 an\u00e1lisis mediante espectrometr\u00eda de masas, estudios de tama\u00f1o y estructura mediante fplc y electroforesis en geles no desnaturalizantes, microscop\u00eda electr\u00f3nica de transmisi\u00f3n para analizar las part\u00edculas reconstituidas y estudio de la estabilidad proteica mediante desnaturalizaci\u00f3n t\u00e9rmica. Los resultados de las distintas t\u00e9cnicas sugieren la existencia de diferencias a nivel estructural. La apo a-i z parece adoptar una conformaci\u00f3n m\u00e1s abierta tanto en forma de prote\u00edna libre como en part\u00edculas rhdl. Asimismo el patr\u00f3n de desnaturalizaci\u00f3n t\u00e9rmica indica que la apo a-i z ser\u00eda menos estable que la forma nativa. la caracterizaci\u00f3n funcional consist\u00eda en el estudio del proceso de eflujo de colesterol desde macr\u00f3fagos, utilizando un marcaje radiactivo, y en el estudio de la captaci\u00f3n de colesterol por parte de una l\u00ednea celular hep\u00e1tica a trav\u00e9s de microscop\u00eda de fluorescencia y citometr\u00eda de flujo. el an\u00e1lisis de la variante apo a-i zaragoza mostr\u00f3 una mayor eficacia en la cesi\u00f3n de colesterol a c\u00e9lulas hep\u00e1ticas y un ligero aumento en el eflujo de colesterol desde macr\u00f3fagos, lo cual contribuir\u00eda a explicar parcialmente sus propiedades
