La acetilaci\u00f3n alfa-aminoterminal est\u00e1 catalizada por las enzimas acetiltransferasas aminoterminales (nat) que transfieren el grupo acetilo desde el acetil coenzima a (acetil coa) al grupo alfa-amino de los residuos aminoterminales de las prote\u00ednas. Se trata de un proceso irreversible que ocurre cotraduccionalmente en el 85% de las prote\u00ednas eucariotas.   \tel complejo enzim\u00e1tico humano natb (hnatb) est\u00e1 constituido por la subunidad catal\u00edtica hnaa20p y la subunidad auxiliar hnaa25p. Estas dos prote\u00ednas interaccionan para formar un complejo estable que se ha conservado desde levaduras hasta humanos tanto en su composici\u00f3n como en la especificidad de sustratos que acetilan. La realizaci\u00f3n de mutantes de deleci\u00f3n de la prote\u00edna hnaa20p ha permitido delimitar la zona de interacci\u00f3n con la subunidad auxiliar hnaa25p a sus 105 primeros amino\u00e1cidos.   \tlas subunidades del complejo hnatb se encuentran asociadas al ribosoma tanto en humanos como en levaduras. No obstante, en humanos se ha detectado una gran parte de sus subunidades no asociadas al ribosoma. Mediante an\u00e1lisis de fraccionamiento subcelular de c\u00e9lulas hela se ha determinado la localizaci\u00f3n de las prote\u00ednas hnaa20p y hnaa25p tanto en el n\u00facleo como en la fracci\u00f3n citos\u00f3lica.  \tla caracterizaci\u00f3n de la actividad enzim\u00e1tica del complejo hnatb mediante ensayos de acetilaci\u00f3n aminoterminal in vitro demostr\u00f3 que hnatb acetila la metionina aminoterminal de sustratos cuyos dos primeros residuos son met-asp y met-glu, tal y como hab\u00eda sido descrito en levaduras.
