Hemos utilizado la linea celular k562, derivada de una leucemia mieloide cronica (lmc) humana, como modelo para estudiar la interaccion funcional entre c-myc y p53. Utilizando dos metodos diferentes, generamos dobles transfectantes k562 con expresion condicional para c-myc y p53. Las celulas transfectadas expresan p53val135 mutante, que adopta una conformacion silvestre a 32\u00c2\u00bac, mientras que la induccion de c-myc fue aumentada con un vector de expresion inducible por zn. Hemos visto que con la conformacion silvestre de p53 resulta en una parada del ciclo celular y apoptosis en k562, pero que c-myc es capaz de rescatarlas de las apoptosis. La inhibicion de la apoptosis fue determinada por estudios morfologicos, union de anexina v, actividad metabolica y fragmentacion internucleosomal del dna. Ademas myc no interfiere con la localizacion y degradacion de p53. Por otro lado, la expresion de myc inhibe la transactivacion mediada por p53 de varios gees diana de p53 como mdm2, bax, p21waf1, pg13, 14-3-3, perp pero no de ciclina g. Para estudiar si c-myc afectaba a p53 a nivel transcripcional, contrasfectamos el gen indicador luciferasa que lleva el promotor del gen diana de p53 a estudiar, con un vector de expresion c-myc. Ademas el cambio a 32\u00c2\u00bac va acompa\u00f1ado de una bajada de la actividad quinasa de ciclina e\/cdk2, medida mediante un ensayo quinasa. Los datos son consistentes con la sobreexpresion de c-myc en presencia de p53 silvestre observada en la mayoria de las lmc en crisis blastica.
