Dnak de escherichia coli es la chaperona hsp70 mejor caracterizada, que junto con dnaj (hsp40) y el intercambiador de nucle\u00f3tido grpe constituyen el sistema dnak. En esta tesis se estudia la interacci\u00f3n entre dnaj y sustratos que poseen diferentes propiedades conformacionales y se analiza la funci\u00f3n de los diferentes dominios de la chaperona en esta interacci\u00f3n, destacando la importancia del dominio rico en g\/f de dnaj en la interacci\u00f3n con sustratos nativos y parcialmente plegados. Adem\u00e1s, se caracteriza la formaci\u00f3n de complejos entre el sustrato nativo repe y las chaperonas dnak y dnaj y se estudia su influencia sobre el estado oligom\u00e9rico y la funci\u00f3n biol\u00f3gica del sustrato. Por \u00faltimo, se pretende estimar la energ\u00eda que puede proporcionar el sistema dnak para modificar la conformaci\u00f3n de repe, ya que dnak es una m\u00e1quina molecular que consume atp para llevar a cabo su funci\u00f3n. Para lograr este objetivo se han dise\u00f1ado diferentes mutantes de repe cuya interfase dim\u00e9rica es m\u00e1s estable que la de la prote\u00edna silvestre.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
