Esta v\u00eda est\u00e1 regulada por varias familias deantagonistas entre las que se encuentran las prote\u00ednas relacionadas con la familia de frizzled (sfrp) y lafamilia de dickkopf (dkk). Dkk-3 es un mediador que regula indirectamente la de la v\u00eda de wnt. En los\u00faltimos a\u00f1os su funci\u00f3n se ha asociado a la regulaci\u00f3n de la morfog\u00e9nesis. Esta tesis se centra en elestudio de los mecanismos de regulaci\u00f3n y de la funci\u00f3n de dkk-3 en la diferenciaci\u00f3n neuronal tomandocomo modelo experimental la l\u00ednea de c\u00e9lulas madre de teratocarcinoma ntera-2 (nt2). Losexperimentos realizados en esta tesis han demostrado que durante la diferenciaci\u00f3n inducida por \u00e1cidoretin\u00f3ico se produce un incremento de la expresi\u00f3n de dkk-3. Los experimentos de silenciamiento g\u00e9nicomediante el uso de shrnas revelaron que la disminuci\u00f3n de la expresi\u00f3n end\u00f3gena de dkk-3 produc\u00edauna inhibici\u00f3n de la diferenciaci\u00f3n neuronal en c\u00e9lulas cultivadas en presencia de \u00e1cido retin\u00f3ico.Tambi\u00e9n se observaron cambios a nivel de adhesi\u00f3n celular en las c\u00e9lulas transfectadas con dkk-3shrna. Adem\u00e1s el estudi\u00f3 de modelos de cultivos celulares tridimensionales mostr\u00f3 que la disminuci\u00f3nde la expresi\u00f3n end\u00f3gena de dkk-3, que fueron analizados mediante m\u00e9todos de an\u00e1lisis de im\u00e1genesmicrosc\u00f3picas, daba lugar a una inhibici\u00f3n del efecto del \u00e1cido retin\u00f3ico en las neurosferas derivadas dec\u00e9lulas transfectadas con shrna. As\u00ed mismo, con el fin de estudiar el efecto de la expresi\u00f3n ect\u00f3pica dedkk-3, se llev\u00f3 a cabo la purificaci\u00f3n de la prote\u00edna recombinante a partir de medio condicionado dec\u00e9lulas hek293 que expresaban de manera constitutiva dkk-3, pero la cantidad de prote\u00edna obtenida fueinsuficiente a cabo estudios funcionales. Por esta raz\u00f3n, se estudi\u00f3 el efecto de la sobreexpresi\u00f3n de dkk-3 en la v\u00eda de se\u00f1alizaci\u00f3n de jak\/sta t, que ha sido postulada como una de las posibles v\u00edas dese\u00f1alizaci\u00f3n sujetas a una regulaci\u00f3n directa por dkk-3. Los resultados obtenidos en esta tesis relacionanla funci\u00f3n de dkk-3 con la diferenciaci\u00f3n inducida por \u00e1cido retin\u00f3ico en las c\u00e9lulas nt2.
