La proliferaci\u00f3n celular por su parte, requiere de una estricta regulaci\u00f3n transcripcional de un gran n\u00famero de genes, de manera que la expresi\u00f3n aberrante de un \u00fanico regulador transcripcional puede resultar en una proliferaci\u00f3n celular defectuosa y alteraciones en la homeostasis tisular. Se ha descrito previamente que los factores de transcripci\u00f3n e2f1 y e2f2 comparten un papel fundamental en la homeostasis pancre\u00e1tica. As\u00ed, la inactivaci\u00f3n simult\u00e1nea de ambos resulta en la disfunci\u00f3n del p\u00e1ncreas exocrino, desarrollo de diabetes y atrofia de la gl\u00e1ndula pancre\u00e1tica. El objetivo de este trabajo ha sido dilucidar el mecanismo por el que los factores de transcripci\u00f3n e2f1 y e2f2 regulan la homeostasis pancre\u00e1tica, y m\u00e1s concretamente, evaluar el papel de la v\u00eda de p53 en dicho proceso.Los resultados obtenidos han demostrado que la inactivaci\u00f3n de e2f1 y e2f2 en el p\u00e1ncreas resulta en hiper-replicaci\u00f3n del dna, incremento de los niveles de expresi\u00f3n de p53 y acumulaci\u00f3n de la histona h2ax fosforilada, caracter\u00edstico de una inducci\u00f3n de la respuesta al da\u00f1o al dna (ddr). La activaci\u00f3n de la v\u00eda de p53 en p\u00e1ncreas carente de e2f1 y e2f2 conlleva la sobreexpresi\u00f3n de dianas transcripcionales de p53 implicados en apoptosis (bax, puma) y en el bloqueo del ciclo celular (p21cip1, gadd45\u00edY). En el presente trabajo se demuestra que p53 juega un papel fundamental en el desarrollo de la diabetes en ratones doblemente knockout para e2f1 y e2f2 (dko). As\u00ed, la eliminaci\u00f3n adicional de p53 en ratones previene la apoptosis mitocondrial, as\u00ed como la atrofia de la gl\u00e1ndula pancre\u00e1tica y de otras gl\u00e1ndulas, y rescata la diabetes dependiente de insulina descrita previamente en los ratones dko.
