En t\u00e9rminos fisico-qu\u00edmicos, los amiloides constituyen el ensamblaje  macromolecular m\u00e1s basico que se auto-propaga. Los priones de levaduras como es sup35p\/[psi ] han servido de instrumento para llegar a conocer las bases moleculares del cambio conformacional en la nucleaci\u00f3n amiloide, los polimorfismos estructurales  y la transmisi\u00f3n de c\u00e9lula a c\u00e9lula. Pero las secuencias amilog\u00e9nicas en levaduras son ricas en aminoacidos q\/n y difieren de las secuencias amilog\u00e9nicas de proteinas involucradas en proteinopatias amiloides cuya naturaleza aminoac\u00eddica es hidrofobi ca. Adem\u00e1s, los priones de levaduras son determinantes epigen\u00e9ticos de fenotipos beneficiosos, no son agentes causantes de proteinopat\u00edas. el laboratorio de rafael giraldo ha descubierto que el dominio wh1 de la prote\u00edna inicidadora de la replicaci\u00f3n  plasm\u00eddica, repa, puede ensamblarse en fibras amiloides in vitro cuando una secuencia espec\u00edfica de adn act\u00faa como factor alost\u00e9rico in vitro. el trabajo realizado en esta tesis doctoral ha contribuido a caracterizar el amiloide repa-wh1 como sistem a modelo para estudiar proteinopat\u00edas. Este ha consistido principalmente en cuatro partes: primeramente, he estudiado el efecto de la expresi\u00f3n en e. Coli de diversas construcciones repa-wh1 fuisionadas a una prote\u00edna fluorescente monom\u00e9rica roja (mc herry). La expresi\u00f3n de wh1(wt) condujo a un patr\u00f3n difuso de fluorescencia en el citoplasma, mientras que wh1(a31v) dio lugar a corp\u00fasculos fluorescentes (part\u00edculas de prote\u00edna agregada) en n\u00famero y con distribuci\u00f3n espacial variados. Un fluor\u00f3foro  con especificidad por los amiloides (bta-1) colocaliza con las inclusiones, confirmando la naturaleza amiloide de \u00e9stas. Adem\u00e1s la agregaci\u00f3n est\u00e1 promovida por dna, tal y como se hab\u00eda encontrado in vitro. Al tomar im\u00e1genes de microscop\u00eda de fluore scencia a intervalos temporales, observamos que en comparaci\u00f3n con c\u00e9lulas que no expresan repa-wh1, las que portan los amiloides expanden sus tiempos de generaci\u00f3n cinco veces y, finalmente, detienen su proliferaci\u00f3n. Repa-wh1 causa pues la primera proteinopat\u00eda amiloide intracelular no infecciosa descrita en procariotas, por lo que puede ser considerado un prionoide sint\u00e9tico. en segundo lugar, he
