Es considerada una enfermedad  rara con una preValencia de 1\/5.000-1\/8.000. La hht est\u00e1 causada, en un 90  de los casos, por mutaciones en los genes endoglina y alk1, originando hht1 y hht2 respectivamente. Ambas prote\u00edna s son componentes del complejo receptor del tgf-?, Expresadas fundamentalmente en c\u00e9lulas endoteliales, e implicadas de una manera importante en angiog\u00e9nesis. La disminuci\u00f3n de los niveles de estas prote\u00ednas conlleva a una se\u00f1alizaci\u00f3n defectuosa de tgf-?, Alterando la expresi\u00f3n de numerosos genes diana y sus funciones, ocasionando una p\u00e9rdida de la red capilar, dando lugar a una dilataci\u00f3n del vaso y como consecuencia una mezcla de sangre arterial y venosa. En la actualidad no existe curaci\u00f3n d e la hht, existiendo medidas paliativas que ayudan a mejorar la calidad de vida. En el presente trabajo se ha buscado f\u00e1rmacos que de una manera u otra puedan disminuir la sintomatolog\u00eda de la hht. Por un lado se han ensayado  f\u00e1rmacos para compensar  la haploinsuficiencia de endoglina y\/o alk1 en los pacientes para una mejora de la se\u00f1alizaci\u00f3n de tgf-? Encontrando que, compuestos como raloxifeno, estriol, n-acetilciste\u00edna o tacrolimus aumentan la expresi\u00f3n de uno de los dos genes o de ambos. Ta mbi\u00e9n se ha realizado el estudio de f\u00e1rmacos antiangiog\u00e9nicos que puedan disminuir la vasculatura anormal  de los pacientes como geniste\u00edna, resveratrol o propranolol, disminuyendo \u00e9stos la tubulog\u00e9nesis in vitro, o la migraci\u00f3n celular necesaria en la angiog\u00e9nesis. Con f\u00e1rmacos que cumplieran estos requisitos se detendr\u00eda la progresi\u00f3n de las telangiectasias en la mucosa nasal y g\u00e1strica y se evitar\u00edan los sangrados, bien mediante normalizaci\u00f3n de la vasculatura o bien por la inhibici\u00f3n de la f ormaci\u00f3n de estos vasos.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral \u00ab<strong>Ensayos
