En la c\u00e9lula ? Pancre\u00e1tica, esta enzima act\u00faa como sensor de glucosa integrando los niveles de glucemia con la secreci\u00f3n de insulina. En el h\u00edgado , su actividad regula el ritmo de acumulaci\u00f3n de gluc\u00f3geno y la producci\u00f3n de glucosa hep\u00e1tica. Mutaciones en el gen gck que incrementan la actividad de la enzima dan lugar a hipoglucemia debido a hiperinsulinismo, mientras que las mutaciones que dis minuyen la actividad de la enzima producen hiperglucemia o diabetes. Debido a su importancia en la homeostasis de glucosa esta enzima es una de las principales dianas de estudio para el desarrollo de nuevas estrategias de terapia antidiab\u00e9tica.  para  profundizar en el conocimiento de los mecanismos de regulaci\u00f3n de la gk humana, se ha realizado la caracterizaci\u00f3n funcional de un grupo de mutaciones identificadas en pacientes con alteraciones en la homeostasis de glucosa. Once de estas mutaciones  hab\u00edan sido asociadas a hiperglucemia familiar y una a hipoglucemia. El estudio funcional ha consistido en analizar el defecto causado por las distintas mutaciones en la estructura proteica y en la cin\u00e9tica enzim\u00e1tica mediante ensayos de actividad i n vitro. Adem\u00e1s, tambi\u00e9n se ha estudiado su efecto sobre la interacci\u00f3n con prote\u00ednas que se asocian a la gk mediante ensayos de pull down y de doble h\u00edbrido en levadura y sobre la localizaci\u00f3n n\u00facleo-citopl\u00e1smica de la enzima mediante microscop\u00eda co nfocal.  los resultados de esta tesis han permitido dilucidar nuevos aspectos de la bioqu\u00edmica de la gk, resaltando la importancia de las caracter\u00edsticas cin\u00e9ticas y estructurales de la glucoquinasa para su funci\u00f3n como sensor de glucosa en la c\u00e9lula  ?
