Para ello se han purificado tanto vesiculas de secrecion como vesiculas de membrana plasmatica invertidas a partir del mutante de secrecion sec 6-4. Purificadas ambas fracciones se comprobo que las principales proteinas implicadas en los procesos de fusion intracelular se encuentran presentes en las mismas con un perfil de distribucion definido. El proceso de fusion entre vesiculas de secrecion y vesiculas de membrana plasmatica invertidas se midio a traves de las propiedades autoatenuables de la sonda lipidica octadecilrodamina, r18. Dicho proceso es dependiente de la temperatura y de la presencia de ciertos cationes en el medio, mostrando ademas, un perfil de saturacion con el tiempo y con cantidades crecientes de membrana diana. Asimismo, por medio de fragmentos fab contra las proteinas sso 1p (t-snare) y snc1p(v-snare) se ha demostrado que ambas proteinas estan directamente implicadas en el proceso de fusion. Por otra parte,tanto la adicion exogena de atp como la adicion de gtp son capaces de estimular el proceso, lo que indica que ambos nucleotidos participan en el mismo.
