Las prote\u00ednas que median tanto la importaci\u00f3n como laexportaci\u00f3n son las carioferinas, prote\u00ednas modulares, flexibles, capaces de reconocersecuencias espec\u00edficas que portan las mol\u00e9culas cargo para su transporte. Algunos deestos cargos son chaperones nucleares, que acompa\u00f1an y protegen a las histonas ensu tr\u00e1nsito desde el citoplasma al n\u00facleo, donde participan en procesos deremodelaci\u00f3n de la cromatina. En esta tesis hemos abordado el estudio,termodin\u00e1mico y estructural, del complejo de importaci\u00f3n formado por el chaper\u00f3nnucleoplasmina con su receptor de transporte, el heterod\u00edmero formado por lasimportinas \u00c2\u00bf y \u00edY. La np, una prote\u00edna homopentam\u00e9rica, es reconocida con altaafinidad por el heterod\u00edmero \u00c2\u00bf\/\u00edY, pudiendo ocuparse todos sus centros de uni\u00f3n, yform\u00e1ndose complejos flexibles, asim\u00e9tricos y modulares, lo que podr\u00eda facilitar sutr\u00e1nsito a trav\u00e9s de los poros nucleares. Tambi\u00e9n hemos observado que la np puedeasociarse simult\u00e1neamente a histonas e importinas, formando complejos definidosque podr\u00edan representar un posible mecanismo de cotransporte de np e histonas aln\u00facleo. La nucleofosmina (npm) pertenece a la misma familia de chaperones dehistonas que la nucleoplasmina. Para desempe\u00f1ar sus funciones, que afectandirectamente al crecimiento celular, la npm es transportada bidireccionalmente entren\u00facleo y citoplasma, es decir, es una \u00c2\u00bfshuttling protein\u00c2\u00bf. La disfunci\u00f3n de esta prote\u00ednaest\u00e1 implicada en diversas patolog\u00edas humanas; en particular, ciertas mutaciones quese correlacionan con la leucemia mieloide aguda, se caracterizan por una localizaci\u00f3ncelular aberrante de la prote\u00edna. En la tesis hemos abordado la interacci\u00f3n de la npmcon la exportina, con lo que globalmente, se ha perseguido profundizar en laimportaci\u00f3n y exportaci\u00f3n de estos chaperones de histonas
