{"id":113935,"date":"2013-11-03T00:00:00","date_gmt":"2013-11-03T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/papel-del-receptor-p2x4-en-la-muerte-microglial-inducida-por-activacion\/"},"modified":"2013-11-03T00:00:00","modified_gmt":"2013-11-03T00:00:00","slug":"papel-del-receptor-p2x4-en-la-muerte-microglial-inducida-por-activacion","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/neurologia\/papel-del-receptor-p2x4-en-la-muerte-microglial-inducida-por-activacion\/","title":{"rendered":"Papel del receptor p2x4 en la muerte microglial inducida por activaci\u00f3n"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Nuria Vazquez Villoldo <\/strong><\/h2>\n<p>El objetivo general de este trabajo de tesis doctoral ha sido estudiar el papel de la se\u00f1alizaci\u00f3n purin\u00e9rgica, en particular la se\u00f1alizaci\u00f3n mediada por el receptor p2x4, en el proceso de activaci\u00f3n microglial tanto en modelos celulares in vitro como en modelos animales in vivo.Para ello nos propusimos los siguientes objetivos espec\u00edficos:1.Caracterizar la expresi\u00f3n de los receptores purin\u00e9rgicos, en particular del receptor p2x4, en la microgl\u00eda in vitro e in vivo, en condiciones basales y de activaci\u00f3n.2.Analizar si el bloqueo del receptor p2x4 inhibe el proceso de activaci\u00f3n microglial in vitro.3.Analizar la muerte microglial inducida por su activaci\u00f3n con lps y el efecto de antagonistas de receptores purin\u00e9rgicos sobre dicha muerte.4.Estudiar el comportamiento de la microgl\u00eda in vivo, en condiciones patol\u00f3gicas de neuroinflamaci\u00f3n y en el modelo animal de la esclerosis m\u00faltiple.5.Estudiar el efecto del tnp-atp y la ivermectina, antagonista y potenciador del receptor p2x4 respectivamente, sobre el comportamiento de la microgl\u00eda in vivo.En su conjunto, en este estudio hemos demostrado que la activaci\u00f3n microglial con lps y la que tiene lugar en la esclerosis m\u00faltiple induce una sobreexpresi\u00f3n del receptor p2x4, y que dicha sobreexpresi\u00f3n regula funciones microgliales como la retracci\u00f3n de procesos y cambio morfol\u00f3gico, la movilidad de la misma, la liberaci\u00f3n de citoquinas como el tnfa y la muerte microglial. La interferencia en dichos procesos, tanto de la activaci\u00f3n, de la muerte microglial o de ambos, mediante el bloqueo del receptor p2x4 con tnp-atp exacerba el cuadro neurol\u00f3gico en la eae, modelo animal de la esclerosis m\u00faltiple.  Por el contrario, la potenciaci\u00f3n del receptor p2x4 con ivermectina atenua el deterioro neurol\u00f3gico en la eae. Estos resultados sugieren que la modulaci\u00f3n de la activaci\u00f3n microglial mediada por p2x4 podr\u00eda tener un papel beneficioso o reparador en el desarrollo de dicha
