Debido a su reproducibilidad y bajo coste, la microscop\u00eda de fuerza de tracci\u00f3n (tfm, por sus siglas en ingl\u00e9s) se utiliza habitualmente para estimar las fuerzas celulares a partir de un gran n\u00famero de part\u00edculas fluorescentes embebidas en un hidrogel que imita las propiedades mec\u00e1nicas de la matriz extracelular. esta tesis presenta una metodolog\u00eda para la cuantificaci\u00f3n de las fuerzas 3d ejercidas por c\u00e9lulas adheridas a la superficie de un gel el\u00e1stico. La metodolog\u00eda propuesta combina nuevas alternativas para el c\u00e1lculo de los desplazamientos de las part\u00edculas fluorescentes con diferentes m\u00e9todos de control de errores. Las t\u00e9cnicas desarrolladas incluyen una etapa de pre-filtrado para mejorar la calidad de las im\u00e1genes adquiridas de las part\u00edculas fluorescentes. El c\u00e1lculo de las deformaciones del gel causadas por la acci\u00f3n de las fuerzas celulares se ha reformulado como un proceso de registro de imagen. Las fuerzas se han recuperado de una forma eficiente operando en el dominio de fourier, y se han aplicado diferentes m\u00e9todos de control de errores que se adaptan a cada distribuci\u00f3n de fuerza. Por \u00faltimo, se ha presentado un simulador de tfm que reproduce fielmente las condiciones de un experimento de tfm real. El simulador permite  evaluar el rendimiento de los algoritmos actuales y futuros utilizados en tfm bajo una gran variedad de condiciones, siendo una herramienta valiosa para interpretar correctamente los resultados de un experimento de tfm real y distinguir los errores inherentes producidos por los algoritmos de los resultados biol\u00f3gicos reales. Finalmente, los m\u00e9todos computaciones desarrollados se han aplicado a experimentos tfm reales para analizar las fuerzas ejercidas por c\u00e9lulas  madre derivadas de tejido adiposo sobre un gel
