Durante la primera parte de este proyecto se llev\u00f3 a cabo un estudio a nivel estructural del dominio c-terminal de la subunidad vps54. Una mutaci\u00f3n autos\u00f3mica recesiva (l967q) en el c-terminal de la prote\u00edna vps54 es responsable del fenotipo del rat\u00f3n wobbler, uno de los modelos animales de degeneraci\u00f3n neuronal espont\u00e1nea mejor caracterizados. Las principales conclusiones de la primera parte del proyecto de tesis fueron: 1) la subunidad vps54 del complejo garp presenta alta similitud estructural con otros factores de amarre, sugiriendo un mecanismo de acci\u00f3n com\u00fan para el proceso de amarre vesicular. 2) la hidrofobicidad y el tama\u00f1o del residuo en posici\u00f3n 967 son esenciales para la estabilidad y plegamiento de la prote\u00edna. 3) la mutaci\u00f3n causante del fenotipo wobbler (l967q) provoca una mayor velocidad de degradaci\u00f3n tanto in vitro como in vivo. Los niveles reducidos de vps54 condicionan el ensamblado del complejo garp y afectan a los niveles globales del mismo. La segunda parte de la tesis consisti\u00f3 en caracterizar estructural y funcionalmente la interacci\u00f3n entre garp y prote\u00ednas snares para determinar los motivos estructurales que aseguran la especificidad de dicha uni\u00f3n. Las principales conclusiones de la segunda parte del proyecto de tesis fueron: 1) por primera vez, se ha podido caracterizar a nivel molecular y biof\u00edsico el motivo estructural que define la interacci\u00f3n entre la sintaxina 6 y la prote\u00edna de amarre ang2. 2) el motivo estructural de interacci\u00f3n est\u00e1 conservado en otro miembro de la familia de las sintaxinas, la sintaxina 10, abriendo la posibilidad a un patr\u00f3n com\u00fan de interacci\u00f3n entre snares y prote\u00ednas de amarre.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
