Sin embargo, todos ellos se basan en animales modificados gen\u00e9ticamente que presentan algunos inconvenientes tales como una escasa eficiencia de cr\u00eda y enfermedades transg\u00e9n-asociados. Nuestra hip\u00f3tesis se bas\u00f3 en generar un nuevo da\u00f1o hep\u00e1tico inducido por ganciclovir (gcv) despu\u00e9s de la transducci\u00f3n del h\u00edgado del rat\u00f3n con un adenovirus que expresa timidina quinasa (ad-tk) y que permitir\u00eda un injerto eficiente de los hepatocitos trasplantados y por consiguiente una repoblaci\u00f3n del h\u00edgado donante, llegando a reemplazar el par\u00e9nquima anfitri\u00f3n de manera significativa. Nuestros resultados mostraron que el 45% del par\u00e9nquima hep\u00e1tico del rat\u00f3n pod\u00eda ser reemplazado en un trasplante alog\u00e9nico con hepatocitos de rat\u00f3n, mientras que un 25% del par\u00e9nquima era reemplazado en el caso del trasplante de hepatocitos humanos. Nuestros resultados tambi\u00e9n muestran que algunos n\u00f3dulos de hepatocitos humanos presentes en los h\u00edgados quim\u00e9ricos se caracterizan por esteatosis y una menor cantidad de endotelio, mientras que los n\u00f3dulos de hepatocitos de rat\u00f3n alog\u00e9nico son normales.  las c\u00e9lulas madre pluripotentes inducidas (ipscs) se han propuesto como una fuente alternativa a los hepatocitos primarios. En este estudio se generaron ipscs y se diferenciaron hacia hepatocitos. Adem\u00e1s, estas c\u00e9lulas diferenciadas fueron capaces de injertar en h\u00edgados de ratones con da\u00f1os adtk \/gcv. Al final del proceso de diferenciaci\u00f3n, ipscs fueron capaces de expresar algunos marcadores propios de hepatocitos y en el trasplante de c\u00e9lulas diferenciadas durante 15- y 20-d\u00edas se comprob\u00f3 la presencia de c\u00e9lulas similares a hepatocitos. Este estudio aumenta el conocimiento sobre ratones quim\u00e9ricos con trasplante de hepatocitos humanos y nuevas terapias para enfermedades hep\u00e1ticas basadas en c\u00e9lulas
