A pesar del gran avance en el conocimiento sobre las actividades biosint\u00e9ticas y la estructura de los componentes de la pared celular, la informaci\u00f3n disponible sobre las actividades de entrecruzamiento entre los componentes de la pared celular era escasa. El objetivo general de este trabajo ha sido la caracterizaci\u00f3n del papel de las prote\u00ednas crh en la biog\u00e9nesis de la pared celular y la morfog\u00e9nesis de s. Cerevisiae. Gracias al desarrollo de t\u00e9cnicas y aproximaciones experimentales espec\u00edficas e innovadoras hemos descubierto que estas enzimas son las responsables del enlace entre la quitina y el \u00edY-1,3 glucano y el \u00edY-1,6 glucano de la pared celular (cabib et al., 2007; cabib et al., 2008). Hemos caracterizado las propiedades bioqu\u00edmicas de la reacci\u00f3n enzim\u00e1tica que llevan a cabo estas prote\u00ednas in vivo e in vitro (blanco et al., 2013b). Con el objetivo de profundizar en el mecanismo de acci\u00f3n de estas prote\u00ednas en el ensamblaje de los componentes de la pared celular, hemos llevado a cabo un estudio estructura-funci\u00f3n de los diferentes dominios implicados en la cat\u00e1lisis. Los resultados obtenidos han proporcionado informaci\u00f3n relevante y detallada sobre el papel catal\u00edtico de determinados amino\u00e1cidos en la reacci\u00f3n de transglicosilaci\u00f3n llevada a cabo por estas prote\u00ednas (blanco et al., 2013a). Adicionalmente, hemos estudiado la relevancia del enlace entre el glucano y la quitina en la morfog\u00e9nesis de la levadura. Los resultados obtenidos de estudios fenot\u00edpicos realizados sobre mutantes que presentan un defecto en morfog\u00e9nesis, junto a la ausencia del enlace entre la quitina y el glucano (crh1\u00c2\u00bf crh2\u00c2\u00bf), han revelado un papel en regulaci\u00f3n de la morfog\u00e9nesis celular hasta ahora desconocido, llevado a cabo a nivel molecular por el enlace qu\u00edmico entre la quitina y el glucano (blanco et al., 2012). Este control lo realizar\u00eda mediante la restricci\u00f3n del crecimiento en la zona del cuello, ya que la quitina unida al \u00edY-1,3 glucano, evita la remodelaci\u00f3n del mismo mediante su uni\u00f3n al \u00edY-1,6 glucano y a las manoprote\u00ednas. Esta premisa se encuentra avalada por la presencia mayoritaria de quitina unida a \u00edY-1,3 glucano en esta zona, frente a quitina unida al \u00edY-1,6 glucano en la pared lateral y por la existencia de dos poblaciones de \u00edY-1,3 glucano: una sin remodelar y polidispersa, y otra remodelada y de alto peso molecular unida a la quitina (cabib et al., 2012).
