En particular, el an\u00e1lisis del metabolismo celular ha permitido diagnosticar enfermedades, as\u00ed como el desarrollo de nuevos f\u00e1rmacos y tratamientos o la optimizaci\u00f3n de distintos procesos biotecnol\u00f3gicos. Actualmente, la caracterizaci\u00f3n de los distintos procesos metab\u00f3licos y sus alteraciones despierta un gran inter\u00e9s por parte de la comunidad cient\u00edfica. La irrupci\u00f3n de distintas tecnolog\u00edas de alta resoluci\u00f3n molecular, tambi\u00e9n conocidas como -\u00f3micas, junto con aproximaciones hol\u00edsticas, como lo es el modelado basado en restricciones, constituyen una estrategia prometedora a la hora de responder a preguntas biol\u00f3gicas relacionadas con el metabolismo. Sin embargo, las metodolog\u00edas actuales que integran datos procedentes de las tecnolog\u00edas -\u00f3micas con redes metab\u00f3licas a escala gen\u00f3mica est\u00e1n en proceso de desarrollo. El objetivo de esta tesis doctoral es precisamente desarrollar nuevos modelos matem\u00e1ticos que incorporen datos -\u00f3micos en el contexto de redes metab\u00f3licas a escala gen\u00f3mica aprovechando la versatilidad inherente y la eficiencia de los modelos basados en programaci\u00f3n lineal. los modelos matem\u00e1ticos desarrollados en el marco de esta tesis son de naturaleza variada y han sido aplicados a una diversidad de cuestiones m\u00e9dicas y biol\u00f3gicas. las contribuciones m\u00e1s relevantes se detallan a continuaci\u00f3n: \u00c2\u00bf\tse presenta un estudio de genes esenciales para c\u00e9lulas cancer\u00edgenas basado en la t\u00e9cnica \u00abflux balance analysis\u00bb. \u00c2\u00bf\tse propone un nuevo m\u00e9todo para el an\u00e1lisis de los experimentos de isotopos marcados. \u00c2\u00bf\tse presentan dos estrategias para el c\u00e1lculo de modos elementales de flujo en redes de escala gen\u00f3mica. \u00c2\u00bf\tse introducen los carbon flux paths y se extiende su definici\u00f3n a nivel at\u00f3mico. \u00c2\u00bf\tse desarrollan dos modelos matem\u00e1ticos para la integraci\u00f3n de los carbon flux paths con datos
