Se cree que el equilibrio correcto de estos procesos se rige por cambios en el contenido de subunidades de receptores de glutamato de tipo nmda a nivel de sinapsis individuales. Entre \u00e9stos, la expresi\u00f3n de glun3a, subunidad no convencional de receptores nmda, se ha propuesto como un freno molecular en la maduraci\u00f3n sin\u00e1ptica. En este trabajo hemos testado esta hip\u00f3tesis utilizando t\u00e9cnicas de microscop\u00eda confocal para tomar im\u00e1genes de neuronas disociadas y rodajas organotipicas de hipocampo, as\u00ed evaluar el papel de receptores de nmda que contienen glun3a en la din\u00e1mica de las espinas dendr\u00edticas. Nuestros resultados demuestran que la sobreexpresi\u00f3n de glun3a reduce la densidad, aumenta la eliminaci\u00f3n y disminuye la estabilidad de las espinas dendr\u00edticas a trav\u00e9s del tiempo. Por el contrario, el silenciamiento de glun3a reduce la eliminaci\u00f3n de espinas y promueve su estabilidad. El efecto de la sobreexpresi\u00f3n de glun3a se potenci\u00f3 mediante el uso de un mutante glun3a endocitosis-deficiente, y pudo reproducirse mediante silenciamiento de la prote\u00edna adaptadora pacsin1, que impide la endocitosis de glun3a end\u00f3geno. Finalmente, la re-expresi\u00f3n de glun3a en tejido maduro reinstaur\u00f3 niveles juveniles de poda sin\u00e1ptica y estabilidad de espinas. Mecan\u00edsticamente, demostramos que la disminuci\u00f3n de la estabilidad en neuronas que sobreexpresan glun3a puede estar relacionada con una incapacidad del circuito de estabilizar selectivamente espinas dendr\u00edticas en respuesta a protocolos que inducen plasticidad sin\u00e1ptica. En conjunto, estos datos proporcionan una fuerte evidencia de que glun3a impide la estabilizaci\u00f3n sin\u00e1ptica mediada por la actividad promoviendo as\u00ed la poda de las espinas, y sugieren que la expresi\u00f3n de glun3a funciona como una se\u00f1al molecular para el control del \u00c2\u00bftempo\u00c2\u00bf y la magnitud de la maduraci\u00f3n de la
