De entre todos los tipos de epilepsia, la epilepsia del l\u00f3bulo temporal es la m\u00e1s frecuente y la que mayor tasa de resistencia al tratamiento farmacol\u00f3gico presenta. Los mecanismos b\u00e1sicos celulares y moleculares de la epileptog\u00e9nesis est\u00e1n sujetos a investigaci\u00f3n. Hay evidencias que indican que la serotonina estar\u00eda implicada en el proceso de epileptog\u00e9nesis. Esta tesis pretende analizar el proceso de epileptog\u00e9nesis en tres modelos animales y el papel de la v\u00eda seroton\u00e9rgica a trav\u00e9s del receptor 5-ht1a en su desarrollo. El objetivo principal planteado ha sido investigar si el 18f-mppf, un radiotrazador pet antagonista del receptor de serotonina 5-ht1a, es un biomarcador del proceso de epileptog\u00e9nesis in vivo y la posible identificaci\u00f3n de una nueva diana terap\u00e9utica para el control y tratamiento de esta enfermedad.En los tres modelos experimentales de epileptog\u00e9nesis utilizados en este trabajo se ha observado un hipometabolismo en la imagen pet con 18f-fdg en los animales que desarrollaron crisis convulsivas generalizadas. En los modelos cr\u00f3nicos, este hipometabolismo se localizaba en la corteza, lo que sugiere cambios permanentes debidos a la epileptog\u00e9nesis. Los animales sometidos a kindling qu\u00edmico con ptz se pudieron diferenciar en dos subgrupos, dependiendo de si alcanzaban el estado de kindling o no. Los animales kindling positivo presentaban una menor actividad metab\u00f3lica cerebral que los animales kindling negativo. Este hipometabolismo ya era visible tras las 5 primeras sesiones y, adem\u00e1s, en este punto temporal, era significativo tambi\u00e9n respecto al grupo control. Esta diferencia temprana puede detectarse mucho antes que aparezcan cambios conductuales en el animal.En los 3 modelos empleados, la imagen pet con 18f-mppf y los estudios autorradiogr\u00e1ficos demuestran un aumento temprano en la densidad del receptor 5-ht1a en respuesta al est\u00edmulo epileptog\u00e9nico.
