Aunque dnmt1 es la enzima que cataliza la reacci\u00c2\u00bfn de metilaci\u00c2\u00bfn despu\u00c2\u00bfs dela replicaci\u00c2\u00bfn, se encontr\u00c2\u00bf que uhrf1 es la prote\u00c2\u00bfna que realmente reconoce sitioscpg hemi-metilados. Uhrf1 tambi\u00c2\u00bfn puede enlazar con el dominio n-terminal dednmt1 y, por lo tanto, es capaz de jugar un papel en la carga correcta de dnmt1 a lossitios cpg. Sin embargo, no se conoce el mecanismo molecular responsable paradistinguir entre sitios hemi-metilados y no metilados o totalmente metilados por uhrf1.Esta tesis supone una contribuci\u00c2\u00bfn a dicha cuestiones a trav\u00c2\u00bfs simulaciones dedinamica molecular.La primera parte del proyecto de tesis fue para dilucidar el mecanismo f\u00c2\u00bfsico por el cualel dominio sra de uhrf1 es capaz de discriminar entre la uni\u00c2\u00bfn a un adn hemimetiladofrente a la uni\u00c2\u00bfn a un adn no metilado o totalmente metilado, mediante elc\u00c2\u00bflculo de diferencias en las constantes de uni\u00c2\u00bfn a trav\u00c2\u00bfs el concepto de ciclotermodin\u00c2\u00bfmico utilizando el m\u00c2\u00bftodo de integraci\u00c2\u00bfn termodin\u00c2\u00bfmica.En la segunda parte de la tesis, se ha estudiado la propensi\u00c2\u00bfn de \u00c2\u00bfbase flipping\u00c2\u00bf de labase citosina en diferentes estados de metilaci\u00c2\u00bfn en los sitios cpg de un adn.Tambi\u00c2\u00bfn se investig\u00c2\u00bf el proceso de \u00c2\u00bfbase flipping\u00c2\u00bf para la citosina objetivo, mientrasque la citosina metilada en la cadena complementaria est\u00c2\u00bf unida a el dominio sra deuhrf1. El proceso de \u00c2\u00bfbase flipping\u00c2\u00bf se realiz\u00c2\u00bf construyendo un potencial de mediafuerza mediante restricciones a lo largo de las coordenadas seleccionadas.En la \u00c2\u00bfltima parte de la tesis hemos dilucidado el mecanismo fisico de la afinidad deuni\u00c2\u00bfn de mbd1 a di-metilados cpg sitios, mediante el c\u00c2\u00bflculo de diferencias de energ\u00c2\u00bfalibre a trav\u00c2\u00bfs el concepto de ciclo termodin\u00c2\u00bfmico, utilizando la t\u00c2\u00bfcnica de integraci\u00c2\u00bfntermodin\u00c2\u00bfmica.
