En esta tesis doctoral mostramos que las c\u00e9lulas precursoras neurales y perivasculares aisladas de dermis se originan, al menos en parte, a partir de la desdiferenciaci\u00f3n de c\u00e9lulas de schwann y pericitos terminalmente diferenciados, y que su competencia neural viene determinada por los niveles de expresi\u00f3n de sox2. Adem\u00e1s, su modulaci\u00f3n permite la interconversi\u00f3n entre ambos tipos celulares. Este mecanismo es extrapolable a otros \u00f3rganos (como el coraz\u00f3n) y especies, por lo que podr\u00eda reflejar un fen\u00f3meno m\u00e1s generalizado. Por otro lado, y en lo que respecta a aplicaciones cl\u00ednicas, se ha demostrado que las c\u00e9lulas precursoras d\u00e9rmicas presentan capacidad de mielinizaci\u00f3n in vitro mediada por los niveles de expresi\u00f3n de sox2. Tras su aislamiento y purificaci\u00f3n, las c\u00e9lulas precursoras p75ntr+cd56+ podr\u00edan ser firmes candidatas para terapias de reemplazo celular en enfermedades neurodegenerativas. Por \u00faltimo, se ha evaluado en cl\u00ednica el trasplante de miniinjertos de piel con fol\u00edculos pilosos -a los cuales se asocian estas poblaciones celulares- como alternativa terap\u00e9utica para la cicatrizaci\u00f3n de las \u00falceras cr\u00f3nicas cut\u00e1neas, demostr\u00e1ndose la factibilidad de un procedimiento que es seguro, de bajo coste y relativamente sencillo, siendo adem\u00e1s adaptable a la atenci\u00f3n ambulatoria.Abstract the dermis contains numerous multipotent cell populations whose physiological role includes the generation of progeny involved in tissue homeostasis and wound healing. The experimental work presented here shows that neural and perivascular precursor cells from dermis originate, at least in part, from dedifferentiation of terminally differentiated schwann and pericytes, their neural competence being dictated by sox2 expression levels. Further, their modulation allows for interconversion of both cell types. This mechanism is extrapolated to other organs (heart) and species, and may thus reflect a widespread phenomenon. With regard to clinical application, dermal precursor cells mielinate axons in vitro in a process dependent on sox2 levels. Once isolated and purified, p75ntr+cd56+ precursor cells could be promising candidates for cell replacement approaches in neurodegenerative disease. Finally, hair follicle minigraft transplantation \u00c2\u00bf hair follicles are intimately associated with these cell subpopulations- has been shown to be a viable therapeutic alternative for chronic leg ulcer healing.
