El objetivo principal de esta tesis fue determinar si sp a y amps act\u00faan de una manera sin\u00e9rgica para incrementar el potencial efecto bactericida que tienen estos componentes por separado.Los resultados indican que in vitro la sp a fue capaz de actuar sin\u00e9rgicamente con amps end\u00f3genos  beta defensinas o sp bn  y ex\u00f3genos  polimixina b y colistina  frente a diferentes pat\u00f3genos respiratorios  klebsiella pneumoniae k2, pseudomonas aeruginosa o1 y haemophilus influenzae no tipificado  que son resistentes a la sp a. La sp a y un derivado de la polimixina b  pmbn , que no tiene actividad bactericida, son capaces de matar conjuntamente a estos pat\u00f3genos respiratorios. El mecanismo molecular por el que este proceso ocurre es mediante la formaci\u00f3n de complejos proteicos macromoleculares que facilitan la uni\u00f3n de la sp a y amps a la bacteria, perturbando la membrana externa. Estudios con modelos de la membrana externa de bacterias gram negativas nos han permitido demostrar que el complejo sp a pmbn pero no cada factor por separado es capaz de perturbar y permeabilizar dicha membrana.Por otra parte, mediante estudios in vivo, hemos demostrado que el efecto protector del tratamiento conjunto de sp a y sp bn frente a k. Pneumoniae k2 fue mucho mayor que la administraci\u00f3n intratraqueal de cada una de las prote\u00ednas por separado. La acci\u00f3n cooperativa de sp a sp bn se debe a: 1  la actividad microbicida de sp a sp bn pero no de cada uno de los factores; 2  el reclutamiento temprano de neutr\u00f3filos en el fluido broncoalveolar inducido por sp a sp bn; y 3  la inducci\u00f3n de una respuesta inflamatoria temprana del hu\u00e9sped. Finalmente hemos demostrado el efecto terap\u00e9utico de sp a sp bn frente a la infecci\u00f3n por k. Pneumoniae k2 mediante su administraci\u00f3n 6h o 24 h despu\u00e9s de la inoculaci\u00f3n del pat\u00f3geno, es decir, cuando la infecci\u00f3n ya estaba instaurada. Los resultados indican que el complejo sp a sp bn disminuye significativamente la carga bacteriana 24 o 48h post infecci\u00f3n. En conjunto, estos resultados demuestran que la prote\u00edna del fluido alveolar sp a aumenta la defensa inmune innata del pulm\u00f3n frente a infecci\u00f3n por bacterias gram negativas pat\u00f3genas al fortalecer la actividad de p\u00e9ptidos antimicrobianos presentes en el del fluido alveolar.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
