La forma i con actividad piruvato quinasa sigue una cinetica hiperbolica con respecto al adp. El mismo comportamiento muestra la piruvato quinasa ii. El magnesio  cuya presencia es esencial para las actividades piruvato quinasa  muestra cooperatividad positiva en su union a las enzimas. La fru 1  6-bisfosfato se comporta como un efector heterotropico positivo de las reacciones catalizadas por las piruvato quinasas  siendo notablemente mayor el efecto sobre la actividad piruvato quinasa i  pero se muestra como un efector alosterico positivo de la piruvato quinasa ii.
