Para ello se ha desarrollado un estudio sobre las herramientas farmacol\u00f3gicas disponibles, centr\u00e1ndonos en algunos donadores de no de car\u00e1cter y propiedades muy diferentes entre s\u00ed. Gracias a este estudio se consiguieron determinar las sustancias idoneas para los estudios desarrollados a continuaic\u00f3n.  seguidamente se procedi\u00f3 al estudio del mecanismo de desensiblizaci\u00f3n a corto plazo de la enzima guanilato ciclasa soluble originada por tratamiento con agentes estimulantes de la v\u00eda. Los resultados demostraron que el proceso conduce a una defosforilaci\u00f3n de la enzima que provoca la incapacidad por parte de la gcs de responder a est\u00edmulos posteriores. Este estado es reversible en el timpo por foforilaci\u00f3n de la prote\u00edna.  finalmente se abord\u00f3 el tema de la tolerancia que se desencadena como consecuencia de tratamientos prolongados con no. El estuido de los mecanismos moleculares responsables de este fen\u00f3meno, que conduce a una disminuci\u00f3n en la capacidad de respuesta a no y, por tanto, a una menor efecto del compuesto, nos llev\u00f3 a estudiar los niveles de expresi\u00f3n de la prote\u00edna y del arnm que codidifca para \u00e9sta. Los resultados indican un complejo mecanismo de regulaci\u00f3n que incluye una modificaci\u00f3n y posterior degradaci\u00f3n de guanilato ciclasa soluble y una disminuci\u00f3n de los niveles de arnm que codifica para la prote\u00edna. Este efecto de modulaci\u00f3n negativa de la v\u00eda est\u00e1 mediado por los segundos mensajeros de la ruta.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
