Carme Espunya Prat <\/strong><\/h2>\n<p>La prote\u00edna quinasa ck2 es una ser\/thr fosfotransferasa que participa en las rutas de transducci\u00f3n de se\u00f1al relacionadas con la proliferaci\u00f3n celular. En su forma m\u00e1s com\u00fan, es una enzima oligom\u00e9rica formada por dos tipos de subunidades, catal\u00edtica (alfa) y reguladora (beta), que se organizan en un tetr\u00e1mero activo del tipo alfa2 y beta2. El objetivo de esta tesis doctoral fue el estudio a nivel bioqu\u00edmico y funcional de la prote\u00edna quinasa ck2 de sistemas vegetales, especialmente en relaci\u00f3n a la proliferaci\u00f3n celular.  se purific\u00f3 por cromatograf\u00eda l\u00edquida la ck2 de plantas de arabidopsis thaliana. En esta especie, como en la mayor\u00eda de plantas, se demostr\u00f3 que coexisten isoformas monom\u00e9ricas (alfa9 con isoformas oligom\u00e9ricas (alfa2beta2), ambas activas enzim\u00e1ticamente y que muestran propiedades bioqu\u00edmicas diferentes.  para el estudio de la regulaci\u00f3n de la ck2 durante la divisi\u00f3n celular, se utiliz\u00f3 como sistema biol\u00f3gico la l\u00ednea celular by-2 de tabaco, que es altamente sincronizable. La expresi\u00f3n global de las subunidades alfa y beta era constitutiva a lo largo del ciclo celular. La actividad enzim\u00e1tica oscilaba a lo largo de ciclo y mostraba picos en la transici\u00f3n g1\/s y en mitosis. Se propone que las pliaminas, los moduladores alost\u00e9ricos m\u00e1s importantes de la actividad ck2, podr\u00edan ser en parte responsables de las oscilaciones de actividad. La inhibici\u00f3n in vivo de la ck2 con el inhibidor espec\u00edfico 4,5,6,7-tetrabromebenzotriazole, corrobor\u00f3 los picos de actividad. el bloqueo del ciclo celular en las fases s y g2 provoc\u00f3 la muerte de las c\u00e9lulas en un intervalo corto de tiempo. El bloqueo en la fase g1 no imped\u00eda que \u00e9stas iniciasen la replicaci\u00f3n en el momento previsto, si bien no eran capaces de completarla porque condensaban prematurament la cromatina.
