La primera hip\u00f3tesis estudiada fue la activaci\u00f3n de los macr\u00f3fagos hep\u00e1ticos como c\u00e9lulas que liberan citocinas implicadas en la activaci\u00f3n de mol\u00e9culas de adhesi\u00f3n en el pulm\u00f3n. Se observ\u00f3 que la lesi\u00f3n pulmonar asociada a la pancreatitis est\u00e1 mediada por la activaci\u00f3n de las c\u00e9lulas de kupffer y que la pancreatitis induce la expresi\u00f3n de p-selectina en el endotelio pulmonar, siendo \u00e9ste un proceso independiente de la activaci\u00f3n de las c\u00e9lulas de kupffer. Se estudi\u00f3 entonces una via alternativa de activaci\u00f3n de la p-selectina.  una fuente importante de radicales libres de ox\u00edgeno (rlo) en la pancreatitis aguda es la enzima xantina oxidasa y se conoce que estos rlo actuan induciendo la expresi\u00f3n de p-selectina en modelos in vitro. En nuestro modelo experimental observamos que el incremento de la expresi\u00f3n de la p-selectina en pulm\u00f3n parece ser disparada por un mecanismo dependiente de los radicals libres generados por la xantina oxidasa liberada por el p\u00e1ncreas.  para estudiar el mecanismo, se evalu\u00f3 la expresi\u00f3n de p-selectina y la s\u00edntesis del factor de activaci\u00f3n plaquetario (paf). Como es conocido que la poli(adp-ribosa) sintetasa (pars) actua como mediador de la activaci\u00f3n de la p-selectina en respuesta a la estimulaci\u00f3n de los rlo, se evalu\u00f3 el efecto de la inhibici\u00f3n de este enzima. Finalmente, agentes que actuan como \u00abscavengers\u00bb del radical super\u00f3xido (super\u00f3xido dismutasa) o del per\u00f3xido de hidr\u00f3geno (catalasa) se utilizaron para identificar el papel de cada componente en la patofisiolog\u00eda de la disfunci\u00f3n pulmonar asociada a la pancreatitis .
