{"id":12751,"date":"2018-03-09T08:58:44","date_gmt":"2018-03-09T08:58:44","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-del-efecto-antifibrogenico-del-tnf-alfa-en-las-celulas-estelares-hepaticas\/"},"modified":"2018-03-09T08:58:44","modified_gmt":"2018-03-09T08:58:44","slug":"estudio-del-efecto-antifibrogenico-del-tnf-alfa-en-las-celulas-estelares-hepaticas","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/estudio-del-efecto-antifibrogenico-del-tnf-alfa-en-las-celulas-estelares-hepaticas\/","title":{"rendered":"Estudio del efecto antifibrogenico del tnf-alfa. en las celulas estelares hepaticas"},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Varela Rey Marta M. <\/strong><\/h2>\n<p>El factor de necrosis tumoral-a(tnf-a), disminuye la expresi\u00f3n del gen de procol\u00e1geno z1(i) en las celulas estelares hepaticas en cultivo (ceh). en este trabajo se ha estudiado tanto el papel del estr\u00e9s oxidativo,como el de la se\u00f1alizacion intracelular en la acci\u00f3n antifibrog\u00e9nica del tnf-a. hemos encontrado que el tnf-a no aumenta en la ceg las especies de oxigeno reactivas y que los antioxidantes no previenen el efecto antifibrogenico de la citoquina. Sin embargo, el tratamiento con tnf-a disminuye la concentraci\u00f3n intracelular de glutation reducido,y este efecto esta implicado en la acci\u00f3n antifibrogenica del tnf-a.  ademas, hemos estudiado, tanto el papel de las esfingomielinasa \u00e1cida (emasa-a),como el de quinasas miembros de la superfamilia de las mapkinasas, concretamente el de la mapk, la sapk y el de la p38 mapk, en la acci\u00f3n antiofibrog\u00e9nica del tnf-a. Hemos encontrado que tras el tratamiento con tnf-a disminuye la fosforilaci\u00f3n de la p38 mapk, y que esta disminuci\u00f3n se correlaciona con la inhibici\u00f3n en los niveles de rnam del procol\u00e1geno 1a(i) debida a la citoquina. Ambos efectos son dependientes de emasa-a, ya que pueden prevenirse con el pretratamiento con el inhibidor sr33557. La implicacion de la p38 mapk en la regulaci\u00f3n de los niveles de rnam de procol\u00e1geno 1a (i) se confirm\u00f3 por el incremento en los niveles de fosforilaci\u00f3n de la p38mapk en respuesta a una citoquina que induce la exprexi\u00f3n del col\u00e1geno en ceh, el tgf-b. El inhibidor de la p38mapk,sb203580, mimetiza el efecto inhibitorio del tnf-a sobre los niveles de rnam de procol\u00e1geno 1a(i) y previene la induccion en los mismos debida al tgf-b.
