Los an\u00e1logos no hipercalc\u00e9micos de la vitamina d3(1a,25(oh)2d3) son drogas con posible acci\u00f3n sobre este tipo de neoplasia. Nosotros mostramos que estos compuestos promueven la diferenciaci\u00f3n de celulas de carcinoma de colon humano, sw480, que expresan el receptor de la vitamina d(vdr) (sw480-adh), mientras que no tienen efecto sobre la subl\u00ednea sw480-r o la l\u00ednea metastasica sw620, que han perdido la expresi\u00f3n de vdr.  1a,25(oh)2d3 induce la expresi\u00f3n de e-cadherina y otras proteinas de adhesion intercelular (ocludina, zo-1,zo-2, vinculina). Este tratamiento tambien provoc\u00f3 el aumento progresivo de la resistencia transepitelial (ter) en las celulas sw480-adh, indicando la adquisicion de uniones ocluyentes funcionales.  la actinomicina d bloque\u00f3 completamente la inducci\u00f3n de e-cadherina, mientras la cicloheximida mostro una fuerte inhibici\u00f3n. Por otro lado, el tratamiento con 1a,25(oh)2d3 indujo 1,8 veces la trascripcion de la construccion con 1,1 kb del promotor del gen humano de la e-cadherina. sin embargo, la vida media del mrna de la e-cadherina no se vio afectada por la 1a,25(oh)2d3, en celulas sw480-adh. Todo ello indica que probablemente la 1a, 25(oh)2d3 induce la expresi\u00f3n de e-cadherina por un mecanismo transcripional que requiere la sintesis de proteinas de novo.  la 1a,25(oh)2d3 promueve la translocacion de b-catenina, placoglobina y zo-1 desde el n\u00facleo y el citoplasma a la membrana citoplasmatica, inhibiendo probablemente asi su actividad transcripcional.  el vdr activado por su ligando es capaz de competir con el tcf-4 por su union a b-catenina. De acuerdo con ello, observamos que el tcf-4 es capaz de boquear la actividad transcripconal del vdr, y que por el contrario,la b-catenina es capaz de potenciar la inducci\u00f3n por 1a, 25(oh)2 d3 de un vdre consenso.
