Auxiliadora <\/strong><\/h2>\n<p>Actualmente se tiene un gran conocimiento, a nivel molecular, de algunas rutas catabolicas microbianas de compuestos aromaticos, especialmente de las pertenecientes al genero pseudomonas los primeros trabajos en el estudio de la capacidad de e. Coli para metabolizar compuestos aromaticos fueron realizados por cooper y skinner (1980). Estos autores establecieron que la estirpe c de e. Coli mineraliza el 3- y 4-hpa por una ruta inducible de tipo meta, donde el homoprotocatecuato actua como intermediario central dihidroxilado. En esta tesis doctoral se ha clonado y caracterizado a nivel molecular la ruta catabolica del 4-hpa de e. Coli w. Esta ruta esta compuesta por once genes; ocho genes estructurales organizados en dos operones catabolicos, el operon hpagedfhi u operon meta y el operon hpabc u operon de la 4-hidroxifenilacetato (4-hpa)- hidroxilasa, dos genes reguladores hpar y hpaa, y el gen hpax de funcion desconocida, que esta relacionado con la superfamilia de transportadores transmembranales mfs. La 4-hpa-hidroxilasa es un sistema enzimatico dependiente de nadh y fad formado por dos componentes proteicos, la proteinas hpab y hpac. La primera es el componente flavoproteico del sistema y la segunda actua como proteina cooperadora del mismo aunque, aparentemente, la formacion del complejo fad-enzima requiere la presencia de ambas. Este sistema enzimatico presenta un rango muy amplio de especificidad de sustrato ya que ademas de hidroxilar 3- y 4-hpa puede hidroxilar otros compuestos aromaticos. La proteina hpaa es un miembro de familia de reguladores xyis\/arac que regula positivamente la expresion del operon hpabc utilizando como efectores 3-hpa, 4-hpa y pa.
