Aunque estan siendo objeto de activa investigacion todavia se tiene un conocimiento limitado de su funcion. En esta memoria se analiza la funcion de estas proteinas in vivo utilizando ratones transgenicos como herramienta principal, y se demuestra que la alteracion del patron de expresion de queratinas produce profundas alteraciones en la estructura y funcion de la epidermis y otros tejidos no epiteliales. Las graves lesiones que producen en los ratones el efecto de la expresion ectopica de la queratina k10 (algunas muy similares a las que presentan los pacientes de paquioniquia congenita), nos permiten deducir que la queratina k10 juega un papel preponderante en la funcion y diferenciacion epiteliales; las queratinas no son intercambiables, sino que su patron especifico de expresion es necesario para el mantenimiento de la estructura y funcionalidad de los epitelios.
