{"id":129209,"date":"1996-01-01T00:00:00","date_gmt":"1996-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/analisis-de-curvas-de-dilucion-aplicado-al-estudio-del-transporte-de-aminoacidos-en-celulas-perfundidas\/"},"modified":"1996-01-01T00:00:00","modified_gmt":"1996-01-01T00:00:00","slug":"analisis-de-curvas-de-dilucion-aplicado-al-estudio-del-transporte-de-aminoacidos-en-celulas-perfundidas","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/analisis-de-curvas-de-dilucion-aplicado-al-estudio-del-transporte-de-aminoacidos-en-celulas-perfundidas\/","title":{"rendered":"Analisis de curvas de dilucion aplicado al estudio del transporte de aminoacidos en celulas perfundidas."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Antonio Manteca Gonzalez <\/strong><\/h2>\n<p>Se han efectuado medidas de captacion e inhibiciones cruzadas de aminoacidos y de glucosa, en celulas aisladas de adenocarcinoma de colon humano, perfundidas en columna con una solucion que contenia la sustancia en estudio marcada con 14c y una molecula de referencia extracelular marcada con 3h, con el objeto de caracterizar los sistemas de transporte implicados para estas sustancias en celulas tumorales.Los datos obtenidos sugieren que los principales mediadores que intervienen son: para la gln, los sistemas asc y l; para la ser, asc y l; para la lys, el y+; para la phe, el l, o una variante del mismo.Se observo que la incubacion con insulina durante 24 horas aumento la captacion de fenilalanina, y que la incubacion con dexametasona 24 horas disminuyo la captacion de lisina y serina. La deprivacion de glucosa en el medio de perfusion hizo disminuir la captacion de glutamina y de lisina, y aumento la de serina. La deprivacion de aminoacidos durante 24 h del medio de incubacion hizo aumentar la captacion de glutamina.  la captacion de glucosa en las celulas tumorales, mediada por el glut1, no se vio afectada por la accion de la insulina ni de la dexametasona a corto plazo, pero si a las 24 horas.  la presencia de glutamina en el medio hace aumentar el transporte de glucosa, y el transporte de glutamina tambien se ve estimulado por la presencia de glucosa, por lo que se deduce que hay sinergismo en el transporte de estas dos sustancias en celulas tumorales. Se sugiere la hipotesis de que ambas se complementan como aportadoras de energia, atomos de carbono, nitrogeno y oxigeno, y de anillos de pentosa para la sintesis de bases nitrogenadas, necesarias
